摘 要:为探讨10-羟基癸酸(10-hydroxydecanoic acid,10-HDA)对多囊卵巢综合征(polycystic ovary syndrome,PCOS)小鼠的作用,本试验选取21日龄ICR小鼠,通过皮下注射脱氢表雄酮(dehydroepiandrosterone,DHEA)构建PCOS小鼠模型,并给予10-HDA治疗。通过阴道涂片、HE染色、ELISA、荧光定量PCR等方法,研究10-HDA对PCOS小鼠发情周期、卵巢组织形态、生殖激素分泌及氧化应激水平的影响。结果显示,PCOS小鼠经10-HDA处理后,发情周期恢复正常,囊状卵泡数减少,血清睾酮水平降低,卵巢中雄激素合成基因Cyp17a1(cytochrome P450,family 17,subfamily a,polypeptide 1)的表达显著减少。同时,10-HDA可降低PCOS小鼠血清中黄体生成素水平,增加促卵泡激素水平,使两者比率明显降低。10-HDA改善了PCOS小鼠血清中雌激素和孕酮水平,促进抗氧化应激相关基因Nrf2和Foxo1的表达,降低PCOS小鼠体内的氧化应激水平。以上结果表明,10-HDA对PCOS小鼠具有良好的治疗作用。To investigate the effects of 10-hydroxydecanoic acid(10-HDA)on polycystic ovary syndrome(PCOS)mice,21-day-old ICR mice were selected to establish the PCOS model through subcutaneous injection of dehydroepiandrosterone(DHEA),followed by the treatment with 10-HDA.The effects of 10-HDA on estrous cycle,ovarian morphology,hormone secretion,and oxidative stress of PCOS mice were evaluated using vaginal smears,HE staining,ELISA,real-time fluorescence quantitative PCR,etc.The results showed that after administration of 10-HDA into PCOS mice,estrous cycle was restored,and the number of cystic follicles significantly decreased.Meanwhile,10-HAD diminished the serum testosterone(T)level and restricted the expression of androgen synthesis gene Cyp17a1(cytochrome P450,family 17,subfamily a,polypeptide 1)in PCOS mice ovaries.Additionally,10-HDA reduced the level of luteinizing hormone(LH)in the serum form PCOS mice but increased the level of serum follicle-stimulating hormone(FSH)together with the decline for LH/FSH ratio.Moreover,10-HDA improved the levels of estrogen and progesterone in the serum from PCOS mice,promoted the expression of antioxidant stress-related genes Nrf2 and Foxo1,and weakened the oxidative stress in PCOS mice.In conclusion,these results demonstrated that 10-HDA has significant therapeutic effects on PCOS mice.
