摘  要:硒代蛋氨酸(selenomethionine,SeMet)被认为具有良好的抗氧化性能,可以调节机体的多种生物功能,是一种良好的有机硒源。越来越多的证据表明,有机硒对预防霉菌毒素中毒有明显的拮抗作用,因此,本试验选取SeMet,旨在探讨SeMet对黄曲霉素B1(aflatoxin B1,AFB1)致兔肺部组织损伤的保护作用。将50只35日龄家兔分为5组,分别为对照组(基础饲粮)、模型组(0.3 mg/kg AFB1+基础饲粮)、低硒剂量组(0.2 mg/kg SeMet+0.3 mg/kg AFB1+基础饲粮)、中硒剂量组(0.4 mg/kg SeMet+0.3 mg/kg AFB1+基础饲粮)、高硒剂量组(0.6 mg/kg SeMet+0.3 mg/kg AFB1+基础饲粮),试验期共21 d。试验结束后,计算肺脏指数,进行HE染色和Masson染色并检测肺脏的氧化因子和炎性因子的含量。HE染色结果显示,模型组中肺部组织充血严重,毛细血管和支气管周围渗出大量的红细胞,肺间质红细胞浸润,肺泡囊体积缩小,Ⅱ型肺泡细胞萎缩、胞质浓缩、细胞核皱缩,淋巴细胞和巨噬细胞明显增多;Masson染色结果显示,模型组肺脏纤维化明显;饲料中添加SeMet可以改善家兔AFB1中毒引起的肺脏病理损伤。补充SeMet还提高了GSH-Px、SOD和T-AOC的活性,显著降低了MDA、TNF-α、IL-6和IL-1β的含量,有效缓解了AFB1中毒引起的氧化应激和炎症反应。综上所述,饲料中添加SeMet可以通过提高抗氧化酶的活性,降低炎性因子的表达,有效地改善AFB1致家兔肺脏的损伤,0.2 mg/kg SeMet的改善效果最为显著。To investigate the protective effect of selenomethionine(SeMet)on lung tissue injury induced by aflatoxin B_1(AFB_1)in rabbits.Fifty 35-day-old rabbits were divided into 5 groups.They were the control group(basal diet),model group(0.3 mg/kg AFB,+basal diet),low selenium dose group(0.2 mg/kg SeMet+0.3 mg/kg AFB,+basal diet)and medium selenium dose group(0.4 mg/kg SeMet+0.3 mg/kg AFB_1+basal diet)and high selenium dose group(0.6 mg/kg SeMet+0.3 mg/kg AFB_1+basal diet),the experiment lasted for 21 d.After the test,the lung index was calculated,HE staining and Masson staining were performed,and the contents of oxidative factors and inflammatory factors in the lung were detected.HE staining results showed that in the model group,pulmonary tissue congestion was severe,a large number of red blood cells were exhaled around capillaries and bronchus,pulmonary interstitial red blood cell infiltration,alveolar sac volume decreased,typeⅡalveolar cell atrophy,cytoplasm concentration,cell nucleus shrinkage,lymphocytes and macrophages increased significantly.Masson staining showed that the pulmonary fibrosis was obvious in the model group.Dietary SeMet can improve lung pathological injury caused by AFB_1 poisoning in rabbits.The supplementation of SeMet also increased the activities of GSHPx,SOD and T-AOC,significantly reduced the contents of MDA,TNF-α,IL-6 and IL1-β,and effectively alleviated the oxidative stress and inflammation caused by AFB_1 poisoning.In conclusion,dietary addition of SeMet can improve the lung damage of rabbits induced by AFB_1 by improving the activity of antioxidant enzymes and reducing the expression of inflammatory factors,and the improvement of 0.2 mg/kg SeMet is the most significant.
