雏鹅“痛风综合征”存在多种病毒感染

摘　　要：近年在我国肉鹅养殖主要地区发生了一种以内脏器官表面尿酸盐沉积为主要病理特征,并伴有肌肉、关节和皮下尿酸盐沉积的鹅痛风,为探讨其可能存在的非星状病毒病原感染,采用RT-PCR/PCR方法对来自江苏、四川、广东、福建、山东、河北6省的12份临床典型病例进行了常见水禽病毒的检测,包括鹅星状病毒(GoAstV)、禽呼肠孤病毒(ARV)、禽源星状病毒1型(AAstV-1)、禽源星状病毒2型(AAstV-2)、坦布苏病毒(TMUV)、鹅禽流感病毒(GAIV)、鹅圆环病毒(GoCV)、鹅细小病毒(GPV)、鹅腺病毒(GAV)、鹅出血性多瘤病毒(GHPV),并对分离到的毒株JS2001、JS2002、JS2003、SC2001进行透射电子显微镜观察和致死胚胎的组织病理学观察。RT-PCR/PCR结果显示,GoAstV阳性率为100.0%,AAstV1阳性率为41.7%,AAstV2阳性率为58.3%,ARV阳性率为50.0%,TMUV阳性率为16.7%,AIV、GPV阳性率均为8.3%;其中GoAstV+AAstV1+AAstV2三重阳性率为25.0%(3/12),GoAstV+AAstV1/AAstV2+ARV三重阳性率为25.0%(3/12),GoAstV+AAstV1+AAstV2+ARV四重阳性率为8.3%(1/12)。分离病毒的透射电镜图片显示,4个样品均存在直径大小为20~30 nm、无囊膜的球形病毒颗粒;此外,SC2001还存在1种直径大小约为150 nm、有明显可见的衣壳和双层膜结构的病毒颗粒,而JS2003还存在直径大小为100~200 nm的病毒颗粒,在其双层膜表面有大小为10~20 nm规则排列的小颗粒。临床样本致死鹅胚体出血或黄染,局部或全身性水肿,皮下伴有严重的胶冻样液体渗出;致死鹅胚的组织病理切片显示,心脏、肝脏、肾脏出现严重的炎性细胞浸润,肝脏细胞、心肌细胞、肾小管上皮细胞均出现严重的空泡变性坏死。结果提示,当前中国流行的鹅痛风存在多种属鹅星状病毒和其他未知病毒等多病毒感染,或可被称为雏鹅“痛风综合征”,其可能存在的多病毒感染致病机制还有待进一步研究。In recent years,a novel goose gout disease which characterized by visceral urate deposition and accompanied with urate deposition in muscle,joints or subcutaneous has resulted in serious economic loss to the goose-rearing industry in China.In order to explore causative pathogens besides of goose astrovirus(GoAstV)in the tissue homogenate of 12 classical goose gout cases from Jiangsu,Sichuan,Guangdong,Fujian,Shandong and Hebei Provinces in China,the GoAstV,avian orthoreovirus(ARV),avastrovirus-1(AAstV-1),avastrovirus-2(AAstV-2),Tembusu virus(TMUV),avian influenza virus(AIV),goose circovirus(GoCV),goose parvovirus(GPV),fowl adenovirus(FAdV),goose hemorrhagic polyomavirus(GHPV)were detected by RT-PCR/PCR.The morphological characteristics and pathogenicity of four virus strains designated JS2001,JS2002,JS2003 and SC2001 isolated from the 12 cases were identified respectively by transmission electron microscopy and histopathological examination of 10-embryonic day-old goose embryos without maternal antibody.The results of RT-PCR/PCR showed that the positive rates of GoAstV,AAstV1,AAstV2,ARV,TMUV,AIV and GPV were 100%,41.7%,58.3%,50.0%,16.7%,8.3%and 8.3%,respectively.The triple positive rate of GoAstV+AAstV1+AAstV2 and GoAstV+AAstV1/AAstV2+ARV was 25.0%(3/12).The quadruple positive rate of GoAstV+AAstV1+AAstV2+ARV was 8.3%(1/12).The virus particles of JS2001,JS2002,JS2003 and SC2001 were spherical and 20-30 nm in size.In addition,there were also some spherical virus particles with clearly visible capsid and double-layer membrane in SC2001,and other spherical virus particles with double-layer membrane which arranged regularly with 10-20 nm particle were found in JS2003.Following embryo inoculated with JS2001,JS2002,JS2003 and SC2001,all the dead embryos exhibited severe subcutaneous hemorrhage,red or yellow staining,local or systemic edema accompanied by jelly-like fluid exudation under the skin.Histopathological examination revealed severe inflammatory cell infiltration in the liver,heart and kidney,severe vacuolar deg
