摘 要:奥利司他给肥胖病的治疗带来了新的途径,其化学全合成方法是当下的研究热点。综述了奥利司他的化学全合成工艺,奥利司他的合成工艺有以月桂醛、不饱和脂肪酸甲酯以及1,3,5-环己三醇以等为原料的合成路线,其中尤以月桂醛法最受关注,月桂醛由于碳链较长,可以直接进行缩合,大幅度缩短了反应路线,并且该法具有收率较高、原料便宜易得以及操作条件简单的优势,已经开始被国内部分厂家采用,有良好的市场前景,有望取代目前常用的天然Lipstatin还原法。Orlistat has brought a new approach to the treatment of obesity,and its chemical total synthesis method is the current research hotspot.The total synthesis process of orlistat was reviewed.The synthesis routes of orlistat include lauraldehyde,unsaturated fatty acid methyl ester,1,3,5-cyclohexanetriol andβ-ketoester as raw materials,among which lauraldehyde method is the most concerned.Because of its long carbon chain,lauraldehyde can be directly condensed,which greatly shortens the reaction route.Moreover,this method has the advantages of high yield,cheap and easily available raw materials and simple operating conditions,and has been adopted by some domestic manufacturers and has a good prospect.
