摘  要:目的观察果糖摄入对肝癌细胞增殖的影响,并探讨对c-Myc/SLC1A5信号上调的机制。方法培养HepG2肝癌细胞,添加果糖进行干预,分别设置5、10mmol/L为低、高剂量果糖组,设置0mmol/L果糖为空白对照组;应用CCK-8进行细胞活性检测,克隆形成实验进行细胞增殖情况检测,流式细胞术进行细胞周期和细胞凋亡检测;应用荧光定量PCR检测c-Myc和SLC1A5的mRNA的转录情况,蛋白免疫印迹检测c-Myc和SLC1A5的蛋白表达情况,以明确其机制。结果CCK-8细胞增值率及克隆形成实验细胞增殖结果均显示果糖能促进肝癌细胞的体外增殖(高剂量组>低剂量组>空白组),且差异有统计学意义(P<0.05);流式细胞术检测结果显示果糖能抑制肝癌细胞的体外凋亡(高剂量组<低剂量组<空白组),然而G 2/M期的细胞所占比例结果为高剂量组>低剂量组>空白组。c-Myc和SLC1A5 mRNA的相对表达量及蛋白表达结果均为高剂量组>低剂量组>空白组。结论果糖摄入可以促进肝癌细胞的体外增殖,诱导c-Myc/SLC1A5的表达,该结果提示过多的果糖摄入具有促进肝癌发生与发展的潜在可能,其作用机制可能与c-Myc/SLC1A5的激活密切相关。Objective To observe the effect of fructose intake on the proliferation of hepatocellular carcinoma cells and to explore the mechanism of up-regulation of c-Myc/SLC1A5 signaling.Methods Hepatocellular carcinoma cells HepG2 were cultured and supplemented with fructose for intervention.5 and 10 mmol/L were set as low and high dose fructose groups,respectively,and 0 mmol/L fructose was set as blank group.CCK-8 was used to detect cell activity,clone formation assay was used to detect cell proliferation,flow cytometry was used to detect cell cycle and apoptosis.Fluorescence quantitative PCR was used to detect the mRNA transcription of c-Myc and SLC1A5,and Western blot was used to detect the protein expression of c-Myc and SLC1A5 to clarify the mechanism.Results The proliferation rate of CCK-8 cells and the cell proliferation results of clonal formation experiment showed that fructose could promote the proliferation of Hepatocellular carcinoma cells in vitro(high dose group>low dose group>blank group),and showed a statistically significant difference(P<0.05).The results of flow cytometry showed that fructose could inhibit the apoptosis of hepatoma cells in vitro(high dose group<low dose group<blank group),but the proportion of G 2/M stage cells was high dose group>low dose group>blank group.The mRNA and protein expression of c-Myc and SLC1A5 were high dose group>low dose group>blank group.Conclusion Fructose intake can promote the proliferation of hepatocellular carcinoma cells in vitro and induce the expression of c-Myc/SLC1A5.These results suggest that excessive fructose intake may potentially promote the occurrence and development of hepatocellular carcinoma,and its mechanism may be closely related to the activation of c-Myc/SLC1A5.
