摘 要:目的评价五加参蛤蚧精对肺气虚模型大鼠的药效学。方法采用锯末烟熏法复制肺气虚大鼠模型,通过比较各组大鼠的一般活动状态、血气指标、慢性支气管炎相关生化指标、气道组织学特征,评价五加参蛤蚧精对肺气虚的药效作用。结果与模型组相比,五加参蛤蚧精干预后,大鼠的体质量明显增加(P<0.05);血液中血氧分压[p(O_(2))]、血氧饱和度(saturation of blood oxygen,SaO_(2))明显升高(P<0.001),血二氧化碳分压[p(CO_(2))]明显降低(P<0.05),p H有升高的趋势;血清中的内皮素(endothelin,ET)、IL-1β明显降低(P<0.01或P<0.05),TNF-α有降低的趋势;肺部结构受损部位有明显改善。结论五加参蛤蚧精能显著改善肺气虚大鼠活动状态及肺气虚证的低氧血症和高碳酸血症,改善受损肺部结构和肺通气功能,并有明显的抗炎作用,其作用机制可能与调节炎症细胞因子IL-1β、TNF-α和ET的表达相关。OBJECTIVE To evaluate the pharmacodynamics of Wujiashen Gejiejing on rats with deficiency of lung qi.METHODS The rat model of deficiency of lung qi was established by sawdust fumigation.By comparing the general activity state,blood acid-alkali indexes,biochemical indexes related to chronic bronchitis and airway histological characteristics of rats in each group,the pharmacodynamics of Wujiashen Gejiejing on rats with deficiency of lung qi was evaluated.RESULTS Compared with model group,after the Wujiashen Gejiejing intervention,the body weight of the rats significantly increased;the levels of p(O_(2))and SaO_(2)in blood were significantly increased(P<0.001),p(CO_(2))was significantly decreased(P<0.05),and pH had a tendency to increase;the levels of endothelin(ET)and IL-1βin serum were significantly decreased(P<0.01 or P<0.05),and the levels of TNF-αin serum had a decreasing trend.The damaged lung structures were significantly improved.CONCLUSION Wujiashen Gejiejing can significantly improve the activity state and improve hypoxemia and hypercapnia of lung qi deficiency syndrome,improve the damaged lung structure and the function of lung ventilation,and has obvious antiinflammatory effect.Its mechanism may be related to regulating the expression of inflammatory cytokines IL-1β,TNF-αand ET.
