摩罗丹大蜜丸对慢性萎缩性胃炎大鼠的影响

摘　　要：目的研究摩罗丹大蜜丸对慢性萎缩性胃炎大鼠的影响及其作用机制。方法采用五因素造模法建立慢性萎缩性胃炎大鼠模型,16周后将模型大鼠随机分为模型组,摩罗丹大蜜丸低、高剂量组(2.43、4.86 g/kg),维酶素组(0.32 g/kg),每组10只,另以10只正常大鼠作为对照组,各组给予相应剂量药物8周后,记录其一般生理状况,PH计检测大鼠胃液pH值,肉眼结合HE染色观察大鼠胃黏膜组织病理改变,免疫组化法观察大鼠胃黏膜组织胃泌素分泌细胞(G细胞)及生长抑素分泌细胞(D细胞)的变化,ELISA法检测大鼠血清胃蛋白酶原Ⅰ/胃蛋白酶原Ⅱ(PGⅠ/PGⅡ)、TNF-α、IL-6水平。结果与模型组比较,摩罗丹大蜜丸各剂量组以及维酶素组均可降低慢性萎缩性胃炎大鼠胃液pH值(P<0.05),改善胃黏膜组织病变情况,增加大鼠胃组织G、D细胞数量(P<0.05,P<0.01),升高血清PGⅠ/PGⅡ比值,降低IL-6、TNF-α水平(P<0.05,P<0.01)。结论摩罗丹大蜜丸可有效改善慢性萎缩性胃炎大鼠症状,其作用与调节G、D细胞数量及分泌能力,血清PGⅠ/PGⅡ、炎症因子水平有关。AIM To investigate the effects of Moluodan Dami Pills on chronic atrophic gastritis(CAG)rats and their mechanism.METHODS The rat models were randomly divided into the model group,the low-dose group and high-dose Moluodan Dami Pills groups(2.43 g/kg and 4.86 g/kg),and vitamin A group(0.32 g/kg),following the 16 weeks successful induction of CAG by five-factor modeling method,in contrast to another 10 normal rats of the control group.After 8 weeks corresponding administration,the rats of each group had their general physiological status and pH value of gastric juice assessed;their pathological changes of gastric mucosa observed by naked eyes combined with HE staining;their changes of gastrin-secreting cells(G cells)and somatostatin-secreting cells(D cells)in gastric mucosa observed by immunohistochemistry;and their serum levels of pepsinogenⅠ/pepsinogenⅡ(PGⅠ/PGⅡ)ratio,TNF-αand IL-6 detected by ELISA.RESULTS Compared with the model group,the groups intervened with Moluodan Damei Pills and vitamin A displayed lower pH values of gastric juice(P<0.05),improved pathological changes of gastric mucosa,increased G and D cells counts(P<0.05,P<0.01),increased ratio of serum PGⅠ/PGⅡ,and decreased levels of IL-6 and TNF-α(P<0.05,P<0.01).CONCLUSION Moluodan Dami Pills can effectively improve the symptoms of CAG rats through its influence on the number and secretion abilityof G and D cells,the levels of serum PGⅠ/PGⅡratio and inflammatory factors.
