摘  要:为研究铁皮石斛(Dendrobium officinale)应激活化蛋白激酶(stress-activated protein kinase,SAPK)基因家族的功能,综合多种生物信息学技术鉴定铁皮石斛SAPK(DoSAPK)家族成员,分析它们的理化特性、染色体定位、系统进化树、基因结构和启动子顺式作用元件等,并通过荧光定量PCR技术检测它们在不同组织、低温处理及盐胁迫处理后的表达量。结果表明,铁皮石斛有8个SAPK家族成员,分为group I、group II和group III亚家族,分布在7条染色体上,且2对基因间存在复制关系。在同一亚族中的家族成员具有类似的基因结构、保守基序和蛋白质二级结构。DoSAPK家族基因的启动子区域富含大量激素响应和胁迫应答相关的调控元件。不同DoSAPK成员在铁皮石斛各组织中均有不同程度的表达,具有组织表达特异性。在低温和盐胁迫处理下呈现差异化表达,可能受到低温和盐胁迫的调控,特别是DoSAPK1在铁皮石斛逆境应答过程中可能起着重要的作用。本研究为铁皮石斛DoSAPK家族基因及其功能的深入研究提供了参考。To investigate the potential roles of stress-activated protein kinase(SAPK)gene famil members in Dendrobium officinale,we employed multiple bioinformatics methods to identify the members of this family.The physicochemical properties,chromosomal localization,phylogenetic relationship,gene structure,and cis-acting elements of each D.officinale SAPK(DoSAPK)member were analyzed.In addition,their expression profiles in different tissues and under the low-temperature or salt stress treatment were determined by real-time fluorescence quantitative PCR.The results showed that D.officinale carried eight DoSAPK family members,which belonged to three groups(groups I,II,and III).These genes were located on seven chromosomes,and there were two pairs of genes with replication.The DoSAPK members within the same group had similar gene structures,conserved motifs,and secondary structures.The cis-acting elements in the promoter regions of DoSAPK genes included abundant hormone and stress response elements.DoSAPK family members presented tissue-specific expression in D.officinale.Furthermore,they were differentially expressed under the low-temperature or salt stress treatment,which suggested that they might be involved in the responses to low-temperature and salt stress.Intriguingly,DoSAPK1 might play a role in the abiotic stress responses.The results laid a foundation for in-depth study of the members and roles of the DoSAPK gene family.
