摘 要:采用柱前衍生化高效液相色谱法,对罗望子胶中主要单糖组分进行检测。选用Diamonsil-plus C 18-A^(*)色谱柱,用磷酸盐缓冲液-乙腈溶液流动相进行等度洗脱,流速设置1.0 mL/min,柱温35℃,进样量5μL,检测波长245 nm。罗望子胶中主要单糖组分葡萄糖、木糖和半乳糖的比例为52.05%、20.03%和12.17%。数据在各自浓度范围内线性关系良好(R≥0.9992),加样回收率在100.33%~101.34%之间,RSD小于2.0%,检测限在0.0005~0.0009 mg/L之间。综上,该检测方法适用于罗望子多糖胶中单糖组分测定。Pre-column derivatization high performance liquid chromatography was used to simultaneously detect the proportions of main monosaccharides in tamarind polysaccharide gum.The tamarind polysaccharide gum sample was hydrolyzed with sulfuric acid and derived with 1-phenyl-3-methyl-5-pyrazolone(PMP),and then determined by the instrument.The determination was performed on a Diamonsil-plus C 18-A*column with phosphate buffer-acetonitrile mobile phase at a flow rate of 1.0 mL/min.The column temperature was set at 35℃,the injection volume was set at 5μL,and the detection wavelength was set at 245 nm.The results showed that the proportions of glucose,xylose and galactose in tamarind polysaccharide gum were 52.05%,20.03% and 12.17%,respectively,and the linear relationship was good(R≥0.9992)in their respective concentration ranges.The recovery rate of the three main monosaccharides was between 100.33% and 101.34%,RSD was less than 2.0%,and the detection limit was between 0.0005 and 0.0009 mg/L.The results showed that the method had the advantages of simple operation,stable reliability,high precision and accuracy,and was suitable for the determination of monosaccharide components in tamarind polysaccharide gum,which provided a basis for the determination of monosaccharide content in tamarind polysaccharide gum.
