黄曲霉毒素B_(1)胶体金免疫层析试纸条的制备及性能研究

摘　　要：以黄曲霉毒素B_(1)为研究对象,制作出一种可实现对黄曲霉毒素B_(1)含量快速检测的胶体金试纸条。实验采用竞争抑制法,基于试纸条的层析作用,以胶体金为标记物与黄曲霉毒素B_(1)单抗偶联,将黄曲霉毒素B_(1)抗原和羊抗鼠二抗固定到硝酸纤维膜上作为检测线和质控线,制备出可以快速检测黄曲霉毒素B_(1)的免疫层析试纸条。通过优化,试纸条在37℃条件下,分别在5,10,15,20 d后测试,效果没有明显降低,检测灵敏度为20 ng/mL;与磺胺类药物、“瘦肉精”类药物均无交叉反应。该试纸条具有良好的稳定性和特异性,可实现生物样本中黄曲霉毒素B_(1)的快速大量检测。A colloidal gold test strip was developed for the rapid detection of aflatoxin B_(1).Based on the competitive inhibition method,colloidal gold was used as a marker to couple with aflatoxin B_(1)monoclonal antibody,and aflatoxin B_(1)antigen and goat anti-mouse secondary antibody were immobilized on nitrocellulose membrane as detection and quality control line,respectively.The test strips were optimized and tested at 37℃for 5,10,15 and 20 d,without significant decrease in efficacy and sensitivity(20 ng/mL).The test strips have good stability and specificity and can be used for the rapid and bulk detection of aflatoxin B_(1)in biological samples.
