摘 要:目的在成年小鼠模型上,探究Silkpro Revive 1064 nm激光美容仪局部应用后对皮肤胶原再生和色素沉着的作用,为激光美容仪的临床应用及基础研究提供依据。方法SPF级6-8周龄的ICR雄性小鼠5只,每只ICR小鼠背部取3 cmX3cm面积备皮,沿着小鼠脊柱将左右两侧均分为:未处理组和处理组,处理组使用激光美容仪的1档模式隔天1次,共处理6次,未处理组除备皮外不予任何处理。分别应用苏木素-伊红染色、Western Blot等方法检测真皮层厚度、Ⅰ型胶原、Ⅲ型胶原等指标的变化;SPF级6-8周龄的C57BL/6雄性小鼠3只,每只C57BL/6小鼠背部取3 cm×3 cm面积备皮,利用紫外线灯反复照射方法建立小鼠皮肤色素沉着诱导模型。在此模型上,沿着小鼠脊柱将左右两侧均分为:未处理组和处理组,处理组使用激光美容仪的1档模式隔天1次,共处理6次,未处理组除备皮外不予任何处理。分别应用Fontana-Masson染色、Western Blot等方法检测黑色素含量及酪氨酸酶等指标的变化。结果小鼠皮肤的真皮层厚度和胶原容积均显著增加,Ⅰ型胶原和Ⅲ型胶原在转录和蛋白水平的表达均显著增加。另外,小鼠皮肤的色素沉着程度减轻,黑色素含量显著下降,黑色素合成相关调控蛋白的表达显著下降。结论Silkpro Revive 1064 nm激光美容仪具有促进皮肤胶原再生和抑制色素沉着的作用。Objective To explore the effects of Silkpro Revive 1064 nm laser cosmetic instrument on collagen production and ultraviolet light-induced pigmentation in adult mice models and to provide experimental evidence for basic research and clinical application of 1064 nm laser cosmetic instrument.Methods Five SPF ICR male mice aged 6-8 weeks were used.An area of 3 cm×3 cm on dorsal skin of each mouse to prepared for laser application and were divided into untreated(left side)and treated group(right side)along the spine.The treated group was treated with the first level mode of laser cosmetic instrument every other day for a total of 6 times,and the untreated group was not treated except for skin preparation.Hematoxylin-eosin staining was used to assess the histopathological changes and dermal thickness in the skin.Quantitative Real-time PCR and western blot were employed to determine type I collagen and type III collagen at transcription and protein level,respectively.Three SPF C57BL/6 male mice aged 6-8 weeks were used for the pigmentation study.An area of 3 cm×3 cm on dorsal skin of each mouse were prepared and were exposed to UV light repeatedly to establish the induced pigmentation model.In this model,the dorsal skin with pigmentation were divided evenly along the spine into untreated group(left side)and treated group(right side).The treated group was treated with the first level mode of laser cosmetic instrument every other day for a total of 6 times,and the untreated group was not treated except for skin preparation.Fontana-Masson staining and Western Blot were used to detect the changes in melanin content,proteins related to melanin synthesis such as tyrosinase.Results The dermal thickness and collagen fiber were significantly increased,and the expression of type I and type III collagen was significantly increased at the transcriptional and protein levels.In addition,the skin pigmentation of mice was alleviated,the content of melanin was significantly decreased,and the expression of regulatory proteins related t
