摘 要:为探索不同诱导子对生物合成乌头碱产量的影响,本研究在黄曲霉Aspergillus flavus的液体发酵过程中分别加入金属诱导子(Co^(2+)、Ag^(+)和Cu^(2+))和化学诱导子(茉莉酸甲酯、水杨酸和硝普钠),以及酵母提取物、种子提取物和共生细菌发酵液等生物诱导子进行诱导试验。试验结果表明,Ag^(+)、Cu^(2+)和Co^(2+)都对乌头碱的生物合成有促进作用,其中Ag^(+)浓度为50μmol/L时,乌头碱产量与对照组相比提高了10倍。化学诱导子MJ和SA诱导效果显著,其中100μmol/L MJ的加入能显著提高乌头碱的产量,产量为20.74 mg/L,为对照组的11倍。而酵母提取物、种子提取物和共生细菌发酵液对乌头碱的产量诱导效果均不显著。本论文探究了不同诱导子对黄曲霉生物合成乌头碱的诱导作用,进而明确不同诱导子及不同诱导浓度对乌头碱生物合成产量的影响。In order to explore the effect of different inducers on the yield of biosynthesis aconitine,the present study was carried out in the liquid fermentation of Aspergillus flavus by adding metal inducers(Co^(2+),Ag^(+)and Cu^(2+))and chemical inducers(Methyl Jasmonate,MJ,Salicylic Acid,SA and Sodium nitroprusside,SND),as well as YE(Yeast extract),SE(Seed extract)and Bac(Symbiotic bacterial fermentation broth).The results showed that Ag^(+),Cu^(2+)and Co^(2+)all could promote the biosynthesis of aconitine,in which Ag^(+)at 50μmol/L increased the aconitine yield by 10-fold compared with the control.Induction effects of the chemical inducers MJ and SA were significant,in which the addition of 100μmol/L MJ significantly increased the production of aconitine with a yield of 20.74 mg/L,which was 11-fold higher than that of the control,while YE,SE and Bac did not have any significant effect on the biosynthesis of aconitine by Aspergillus flavus.
