摘 要:目的 建立中药黑骨藤HPLC-ELSD指纹图谱并结合聚类分析和主成分分析进行药材质量评价。方法 采用通用型ELSD检测器建立17个不同产地的黑骨藤药材的指纹图谱,采用HPLC-Q-TOF-MS技术对黑骨藤中化学成分进行表征,并通过指纹图谱相似度评价系统、层次聚类分析和主成分分析评价药材质量。结果 指纹图谱共标定了14个共有峰,采用液质联用技术共鉴定了58个化合物。除2批广西产地黑骨藤指纹图谱与对照图谱的相似度为0.663和0.826,其他15批产地的指纹图谱与对照图谱相似度均在0.9以上。指纹图谱相似度评价、层次聚类分析和主成分分析结果互相印证。结论 黑骨藤HPLC-ELSD指纹图谱研究结合聚类分析和主成分分析为该药材的真伪鉴别、质量控制提供了参考。OBJECTIVE To establish the fingerprint of Periploca forrestii Schltr.for quality evaluation.The quality of medicinal materials was evaluated by cluster analysis and principal component analysis.METHODS High performance liquid chromatography-evaporative light scattering detector(HPLC-ELSD) method was used to establish the fingerprints of 17 different habitats of Periploca forrestii Schltr.The chemical constituents of Periploca forrestii Schltr.were characterized by high performance liquid chromatography-quadrupole-time of flight mass spectrometer,and the quality of the medicinal materials was evaluated by fingerprint similarity evaluation system,hierarchical cluster analysis and principal component analysis.RESULTS A total of 14 common peaks were identified in the fingerprint,and 58 compounds were identified by liquid chromatography-mass spectrometry.Except for the similarity of 0.663 and 0.826 between the fingerprints of the two batches of Guangxi origin and the control map,the similarity of the fingerprints of the other 15 batches of origin and the control map was above 0.9.The results of fingerprint similarity evaluation,hierarchical cluster analysis and principal component analysis were mutually confirmed.CONCLUSION The study of the fingerprint of Periploca forrestii Schltr.combined with cluster analysis and principal component analysis provides a reference for the authenticity identification and quality control of this medicinal material.
