摘 要:目的观察子痫前期大鼠后代胎脑前额叶皮质、海马及小脑中小胶质细胞变化情况。方法通过缩窄腹主动脉及双侧子宫动脉,制造子宫胎盘缺血模型(RUPP),检测20.5d孕鼠动脉血压,尿总蛋白及肾脏病理改变。免疫荧光染色和Westernblot检测胎脑前额叶皮质、海马及小脑内小胶质细胞标记分子Ibal的表达。结果动脉血压检测结果显示,RUPP组动脉血压显著高于对照组。HE染色结果显示,与对照组相比,RUPP组肾小球细胞体积增大,肾小球内细胞数目增多。Westernblot结果显示,与对照组相比,RUPP组胎脑前额叶皮质Ibal蛋白表达增多,但是在海马及小脑无明显改变。免疫荧光染色结果显示,与对照组相比,RUPP组胎脑前额叶皮质Iba1数量增多,但是在海马及小脑无明显改变。结论子痫前期导致胎脑前额叶皮质小胶质细胞增生可能是子代神经发育异常的部分原因。Objective To observe the changes of microglia in prefrontal cortex,hippocampus and cerebellum of fetal brain of preeclampsia rats.Methods The abdominal aorta and bilateral uterine arteries were restrained to make a reduced uterine perfusion pressure(RUPP)model.The arterial blood pressure of the pregnant rat at 20.5 days was measured,and the total urine protein and pathological changes in kidneys were examined,immunofluorescence staining and Western blot were used to detect the expression of ionized calcium binding adaptor molecule-1(Ibal)in microglia in the prefrontal cortex,hippocampus and cerebellum.Results The arterial blood pressure test results showed that the arterial blood pressure of the RUPP group was significantly higher than that of the control group.The results of HE staining showed that compared with the control group,the volume of glomerular cells in the RUPP group was increased,and the number of cells in the glomerulus was increased.Western blot results showed that the protein expression of Ibal in the prefrontal cortex of RUPP group embryos was increased compared with those of control embryos,but those in the hippocampus and cerebellum showed no difference.The results of immunofluorescence staining showed that the number of Ibal positive cells in the prefrontal cortex of RUPP group embryos was increased compared with those of control embryos,and those in hippocampus and cerebellum showed no difference.Conclusion The proliferation of microglial cells in the prefrontal cortex of the fetal brain caursed by preeclampsia may be part of the reason for the abnormal neurodevelopment of the offspring.
