摘  要:目的探讨2016—2021年云南省手足口病CVA9型肠道病毒VP1区基因分子进化特征,为手足口病的预防和控制提供科学依据。方法对云南省2016—2021年各州市采集的手足口病核酸检测阳性的病例标本进行病原分型分析,通过细胞培养,富集病毒后进行VP1区基因测序,采用DNA/Star软件进行核苷酸及氨基酸序列比对和基因进化特征分析。用Mega 5.2软件邻位相接法和Kimura 2参数法构建CVA9的基因进化树。结果云南省共分离到6株CVA9病毒株;CVA9毒株分布在4个州市,阳性病例均表现为轻症,年龄分布在7岁及以下年龄段,发病时间段主要集中在1—2月份。其中VP1区基因在11、12、18、84、132等位点氨基酸变化活跃。CVA9的10个基因组(A~J)之间的核苷酸差异率均大于15%,云南省分离的6株CVA9毒株分布在B基因组的G亚型。各CVA9株之间核苷酸、氨基酸差异率分别为15.75%~24.72%、1.39%~2.16%。云南省分离的6株CVA9毒株和国内云南、山东、浙江参考株遗传距离较近。结论2016—2021年引起手足口病的CVA9病毒散在流行,与国内部分省市引起手足口病的毒株共进化、共循环。Objective To study the molecular evolution characteristics of VP1 gene of CVA9 enterovirus,the pathogen of hand foot and mouth disease(HFMD),in Yunnan province from 2016 to 2021,and to provid some sicientific basis for the prevention and control of HFMD.Methods Pathogenic molecular typing analysis was conducted on positive nucleic acid test for hand,foot and mouth case specimens from Yunnan province from 2016 to 2021,the VP1 gene was sequenced after virus enrichment through cell culture,and nucleotide and amino acid sequence alignment and gene evolution characteristics were analyzed using bioinformatics software.The phylogenetic tree of CVA9 was constructed by Mega 5.2 softwere using the neighbor joining method and kimura 2 parameter.Results A total of 6 CVA9 strain was distributed in four prefectures or cities in Yunnan province,and all patients showed mild symptoms,all the patients were 7 years old or younger.The infection time was mainly from January to February.The VP1 region was active in amino acid changes at loci 11,12,18,84 and 132.The nucleotide difference rates between the 10 genomes(A—J)of CVA9 were greater than 15%,and the 6 CVA9 strains isolated in Yunnan province were distributed in G subtype of the B genome.The difference rates of nucleotide and amino acid among CVA9 strains were 15.75%—24.72%and 1.39%—2.16%,respectively.The genetic distance between the isolated strains in Yunnan province and domestic reference strains in Yunnan,Shandong and Zhejiang was relatively close.Conclusion The CVA9 viruses from 2016 to 2021were sporadic,coevolving and circulating with strains in China.
