摘 要:目的对云南省2022年从手足口病(hand,foot and mouth disease,HFMD)病例中分离到的2株埃可病毒11型(echovirus 11,E11)进行分子生物学鉴定并对它们的VP1区基因特征进行分析。方法对2022年云南省手足口病实验室监测网络收集到的手足口病病例粪便标本进行病毒分离,对阳性标本先进行VP4/VP2结合区逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)和基因测序定型,再对VP4/VP2结合区初步定型的标本进行VP1区RT-PCR和测序,用MEGA 7.0软件构建病毒VP1区基因进化树,进行基因特征分析。结果2022年从云南省手足口病病例粪便标本中检测到2株E11病毒,VP1区基因序列同源性分析结果表明,2株病毒最接近3株在欧洲葡萄膜炎(uveitis)病例中分离的病毒。本研究分离的2株病毒与欧洲3株病毒之间核苷酸相似性(nucleotide,nt)和氨基酸(amino acid,aa)同源性分别为84.37%(差异率15.63%)和95.21%(差异率4.79%)。云南2株和E11原始株(Silva)之间的核苷酸相似性为80.03%(差异率为19.97%),氨基酸同源性为93.50%(差异率6.50%),与原型株(Gregory)的核苷酸相似性为76.95%(差异率23.05%),氨基酸同源性为88.02%(差异率11.98%)。本次云南分离的2株E11为D5亚型。结论2022年云南省手足口病实验室检测到的2株E11为D5亚型,是目前云南和整个中国最为流行的基因亚型。应进一步加强对非脊灰肠道病毒(non-polio enteroviruses,NPEVs)和非EV-A71、非CVA16等手足口病中“其他肠道病毒”(other enteroviruses)尤其是E11病毒的分子生物学检测。Objective To identify molecular biological characterization of 2 strains of echovirus 11(E11)isolated from hand,foot and mouth disease(HFMD)cases in Yunnan province in 2022 and analyze their VP1gere features.Methods Virus isolation was conducted for HFMD stool samples collected from the HFMD laboratory surveillance system of Yunnan province in 2022.Reverse-transcription polymerase chain reaction(RTPCR)and sequencing on VP4/VP2 junction region were conducted for virus setrotyping for virus isolation positive samples.RT-PCR and sequencing on VP1 gene were done for specimens initially typed in the VP4/VP2 binding region.The gene evolution tree of the viral VP1 region was constructed using MEGA 7.0 software for phylogenetical analysis.Results Altogether,2 E11 strains were identified from stool samples of HFMD patients in Yunnan province in 2022.Phylogenetical analysis on the VP1 gene showed that these 2 E11 strains were closest to 3 strains isolated from European uveitis outbreak patients.The nucleotide(nt)and amino acid(aa)similarities between Yunnan and European strains were 84.37%(diversity 15.63%)and 95.21%(diversity 4.79%),respectively.Two Yunnan E11 strains were secondly closed to the prime strain Silva with nt and aa similarities of 80.03%(diversity 19.97%)and 93.50%(diversity 6.50%),respectively.Two Yunnan E11 strains was thirdly most closed to the prototype strain Gregory with nt and aa similarities of 76.95%(diversity 23.05%)and 88.02%(diversity 11.98%),respectively.The 2 Yunnan strains belonged to the D5 sub-genotype.Conclusions Two strains of E11 were identified as D5 sub-genotypes from the HFMD surveillance system in 2022 in Yunnan province,this subgenotype is widely distributed in Yunnan and most other provinces in China.Further efforts should be made to strengthen the molecular biology detection of on-polio enteroviruses(NPEVs)and other enteroviruses in HFMD such as non-EV-A71 and non-CVA16 viruses,especially E11 viruses.
