川芎、益母草有效成分配伍对酪氨酸酶活性、黑色素生成的抑制效应研究

摘  要:目的:探索川芎与益母草有效成分配伍对酪氨酸酶活性和黑色素生成的影响。方法:采用左旋多巴氧化法测定川芎水煎液和益母草水煎以及川芎总生物碱和益母草总生物碱不同质量配比对酪氨酸酶活性的抑制作用,筛选出最佳组合进行后续实验。MTT法检测所筛选出的组合对B16细胞存活率的影响。随后实验分为细胞对照组、α-MSH诱导模型组和模型+所筛选出的最佳组合(50、100、200μg·mL^(-1))、模型+阳性组(曲酸,100μg·mL^(-1))。NaOH裂解法和多巴氧化法检测细胞内黑色素含量和酪氨酸酶活性。结果:通过筛选得出川芎总生物碱和益母草总生物碱质量比为1:2的组合效果最佳。药物配伍25、50、100、200、250μg·mL^(-1)对B16细胞无明显抑制作用。与空白组相比,模型组B16细胞中酪氨酸酶活性、黑色素水平增高(P<0.05,P<0.01),各剂量药物干预后,小鼠黑色素瘤细胞B16中酪氨酸酶活性、黑色素水平降低(P<0.05,P<0.01)。结论:川芎总生物碱和益母草总生物碱质量比为1:2的组合对酪氨酸酶抑制效果最佳,且能显著抑制B16细胞内酪氨酸酶活性和黑色素的生成。Objective:To explore the effects of Chuanxiong and Yimucao on tyrosinase activity and melanin production.Method:The L-dopa oxidation method was used to determine the inhibitory effects of decoctions of Chuanxiong and Yimucao,as well as different mass ratios of total alkaloids of Chuanxiong and Yimucao on tyrosinase activity.The best combination was selected for subsequent experiments.The MTT assay was used to detect the effect of the selected combinations on the survival rate of B16 cells.Then the experiment was divided into a blank control group,a α-MSH-induced model group,a mode+the best combination selected(50,100,200μg·mL^(-1)),model+positive group(kojic acid,100μg·mL^(-1)).The NaOH lysis method and the dopa oxidation method were used to detect the intracellular melanin content and tyrosinase activity.Result:After screening,the combination of the total alkaloids of chuanxiong and motherwort with a mass ratio of 1:2 showed the best effect.The concentrations of 25,50,100,200,250μg·mL^(-1)had no significant inhibitory effect on B16 cells.Compared with the blank group,the tyrosinase activity and melanin level in B16 cells of the model group were increased(P<0.05,P<0.01),and the tyrosinase activity and melanin level in B16 cells of the mouse melanoma cells were decreased after the intervention of each dose of drug(P<0.05,P<0.01).Conclusion:The combination of total alkaloids from Chuanxiong and Yimucao with a mass ratio of 1:2 has the best inhibitory effect on tyrosinase and can significantly inhibit the tyrosinase activity and melanin production in B16 cells.
