摘 要:中医药在治疗椎间盘退变所致腰痛疾病中有广泛的应用。为了研究其治疗机制,建立符合疾病发展规律的动物模型是必不可少的基础工作。目前,用于椎间盘退变研究的动物模型可根据造模方式的不同分为自发模型、化学损伤模型、应力改变模型、脊柱失稳模型、结构损伤模型等。其中,结构损伤模型中的纤维环损伤法所构建的动物模型具有成模率稳定、模型可靠、椎间盘退变进展缓慢等优点,因此较适合作为中医药治疗研究的动物模型。然而,目前该造模方式存在损伤器具不统一、进针角度、留针时间以及针刺次数不一致等问题。该文综述了纤维环损伤法构建椎间盘退变动物模型的机制假设、动物选择、复制方法比较、观察指标以及在中医药研究中的应用,旨在为后续相关中医药机制研究提供可行的造模方案参考。Traditional Chinese medicine has a wide range of applications in the treatment of lumbar pain caused by intervertebral disc degeneration.To study its therapeutic mechanism,it is essential to establish animal models that comply with the development pattern of the disease.Currently,animal models used in intervertebral disc degeneration research can be classified into spontaneous models,chemical injury models,stress-induced models,spinal instability models,and structural injury models,among others.Among them,the animal models constructed by the annular fiber injury method in the structural injury model have advantages such as stable modeling rate,reliable model,and slow progression of intervertebral disc degeneration,making them more suitable as animal models for traditional Chinese medicine research.However,the current method of constructing such models has issues such as inconsistent use of injury instruments,needle insertion angles,retention time,and needle puncture frequency.This article reviews the mechanism hypothesis,animal selection,comparison of replication methods,observation indicators,and application in traditional Chinese medicine research of the annular fiber injury method for constructing animal models of intervertebral disc degeneration,aiming to provide feasible model construction references for subsequent studies on the mechanism of traditional Chinese medicine.
