摘  要:目的探究羧胺三唑(CAI)对人嗜酸性粒细胞白血病细胞系(EOL-1)增殖的影响及相关机制。方法以EOL-1细胞系为研究对象,0.625、1.25、2.5、5、10、20和40μmol·L^(-1)CAI处理对数生长期EOL-1细胞24和48 h,CCK-8法检测细胞增殖,计算抑制率和半数抑制浓度(IC_(50));Western blotting法检测CAI对PDGFRα信号通路关键蛋白的影响。结果CAI对EOL-1细胞增殖具有明显抑制作用,表现出量效和时效关系,CAI处理EOL-1细胞24和48 h的IC_(50)分别为4.22和1.82μmol·L^(-1);CAI降低PDGFRα、Stat3、Akt和Erk 1/2的磷酸化水平,但对总蛋白水平没有明显影响。结论CAI呈剂量和时间依赖性地抑制EOL-1细胞增殖,其作用机制可能与抑制PDGFRα信号通路活化有关。Objective To investigate the effects of carboxyamidotriazole(CAI)on the proliferation of human eosinophilic leukemia cell line(EOL-1)and related mechanisms.Methods EOL-1 cell line was used as the study object,and the cells in logarithmic growth phase were treated with different concentrations of CAI(0.625,1.25,2.5,5,10,20,and 40μmol·L^(-1))for 24 h and 48 h.The cell proliferation was detected by the CCK-8 method,and the inhibition rate and IC_(50) were calculated.The effect of CAI on key proteins of PDGFRαsignaling pathway was determined by Western Blotting.Results CAI significantly inhibited the proliferation of EOL-1 cells,showing the dose-response and time-response relationship,and the IC_(50) of CAI on EOL-1 cells for 24 and 48 h were 4.22 and 1.82μmol·L^(-1),respectively.In addition,CAI reduced the phosphorylation levels of PDGFRα,Stat3,Akt,and Erk 1/2,but there was no significant effect on the total protein levels.Conclusion CAI inhibited EOL-1 cell proliferation in a dose-and time-dependent manner,and its mechanism might be related to inhibition the activation of PDGFRαsignaling pathway.
