菝葜通过NLRP3通路对巴豆油诱发痔疮大鼠模型的改善作用

摘　　要：目的 探讨菝葜通过NLRP3通路对巴豆油诱发痔疮大鼠的改善作用及机制。方法 选取40只6周龄SPF级雌雄各半的SD大鼠,按随机数字表法分为对照组、模型组、菝葜低剂量组、菝葜高剂量组和地奥司明片组,每组8只。造模后菝葜低剂量组、高剂量组及地奥司明片组分别以1.58 g/kg、6.3 g/kg、0.122 g/kg灌胃,1次/d,连续1周。末次灌胃24 h后取大鼠直肠组织及血清,苏木精-伊红(HE)染色检测直肠组织病理变化;试剂盒检测血清中乳酸脱氢酶(LDH)、白细胞介素-1β(IL-1β)、白细胞介素-10(IL-10)、白细胞介素-13(IL-13)和白细胞介素-18(IL-18)的含量;蛋白质印迹法(Western Blot)检测直肠组织中NOD样受体热蛋白结构域相关蛋白3(NLRP3)、半胱氨酸蛋白酶-1剪切体(Cleaved Caspase-1)和消皮素D(GSDMD)蛋白的表达。结果 与对照组比较,模型组大鼠直肠组织病理损伤加重,LDH、IL-1β、IL-18、NLRP3、Cleaved Caspase-1和GSDMD含量均明显升高(P<0.05),IL-10和IL-13含量则明显下降(P<0.05)。与模型组比较,菝葜低剂量组大鼠直肠组织病理损伤减轻,IL-18、NLRP3和GSDMD含量均明显下降(P<0.05),Cleaved Caspase-1、LDH、IL-1β、IL-10和IL-13含量无显著变化(P>0.05);与模型组比较,菝葜高剂量组大鼠的直肠组织病理损伤减轻,LDH、IL-1β、IL-18、NLRP3、Cleaved Caspase-1和GSDMD含量均明显下降(P<0.05),IL-10和IL-13含量无显著变化(P>0.05)。结论 菝葜对痔疮大鼠有较好的治疗效果,其具体机制可能与调节NLRP3通路有关。Objective To investigate the effect of improvment and its mechanism of Smilax china L.on croton oil-induced hemorrhoid rats through NLRP3 pathway.Methods Forty SD rats of 6-week-old SPF grade,half male and female,were selected and randomly divided into control group,model group,low-dose Smilax china L.group,high-dose Smilax china L.group and diosmin tablet group,8 in each group.After modeling,the low-dose Smilax china L.group,high-dose Smilax china L.group and diosmin tablet group were given 1.58 g/kg,6.3 g/kg,and 0.122 g/kg by gavage respectively,once a day for 1 week.Twentyfour hours after the last intragastric administration,the rectal tissue and serum of rats were taken.Hematoxylin eosin(HE)staining was used to detect the pathological changes ofrectal tissue;test kits were used to detect the content oflactate dehydrogenase(LDH),interleukin-1β(IL-1β),IL-18,IL-10 and IL-13 in serum;the expressions of NOD-like receptor thermal protein domain associated protein 3(NLRP3),cleaved cysteinyl aspartate specific proteinase-1(Cleaved Caspase-1)and gasdermin D(GSDMD)proteins in rectal tissues were detected by Western Blot.Results Compared with control group,the rectal tissue pathological damage of model group was aggravated,the expressions of NLRP3,Cleaved Caspase-1,GSDMD,LDH,IL-1βand IL-18 were significantly increased(P<0.05),while the contents of IL-10 and IL-13 decreased significantly(P<0.05).Compared with model group,the rectal tissue pathological damage of the low-dose Smilax china L.group was alleviated,the expressions of IL-18,NLRP3 and GSDMD were significantly decreased(P<0.05),there was no significant difference in Cleaved Caspase-1,LDH,IL-1β,IL-10 and IL-13 content(P>0.05).Compared with model group,the rectal tissue pathological damage of the high-dose Smilax china L.group was alleviated,and the expressions of IL-1β,IL-18,NLRP3,Cleaved Caspase-1 and GSDMD were significantly decreased(P<0.05),there were no significant differences in Cleaved Caspase-1,LDH,IL-1β,IL-10 and IL-13 contents(P>0.05).Conclusion Sm
