摘 要:目的:研究蒙古黄芪Astragalus membranaceus(Fisch.)Bge.var.mongholicus(Bge).Hsiao的化学成分及其对高糖诱导的人肾小管上皮细胞炎性损伤的影响。方法:采用多种柱色谱方法对蒙古黄芪根的95%乙醇提取物乙酸乙酯部位进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构;采用CCK-8法检测单体化合物对高糖诱导的HK-2细胞凋亡率的影响,ELISA分析对细胞炎性因子TNF-α和IL-6含量的影响。结果:从蒙古黄芪中分离得到7个化合物,分别鉴定为:鼠李醚-3-O-α-L-吡喃鼠李糖基(1→2)-[6-O-乙酰基]-β-D-吡喃葡萄糖苷(1)、山柰酚-3-O-β-D-吡喃葡萄糖苷(2)、槲皮素-3-O-β-D-吡喃葡萄糖苷(3)、鼠李醚-3-O-β-D-吡喃葡萄糖苷(4)、鼠李醚-3-O-β-D-新橙皮苷(5)、9,12,13-三羟基-10,15-十八碳二烯酸甲酯(6)、甲基-(9S,10R,11E,13R)-9,10,13-三羟基十八碳-11-烯酸(7)。不同剂量的化合物1~5均可不同程度地降低高糖诱导的HK-2细胞凋亡率,化合物1~4可抑制HK-2细胞TNF-α、IL-6水平的升高。结论:其中,化合物1为新的黄酮苷类化合物,化合物6、7为首次从豆科植物中分离得到。化合物1~4能抑制高糖诱导的HK-2炎症反应。Objective:To study the chemical constituents of Astragalus membranaceus var.mongholicus and the effect on the inflammatory damage in human renal tubular epithelial cells induced by high glucose.Methods:The ethyl acetate extract of 95%ethanol extract of Astragalus membranaceus var.mongholicus was isolated and purified by various chromatography,the structures of obtained compounds were identified by physicochemical properties and spectral data.CCK-8 method was used to detect the influence of monomer compounds on the apoptosis rate of HK-2 cells induced by high sugar,ELISA was used to analyze the influence on the contents of TNF-αand IL-6.Results:Seven compounds were isolated from Astragalus membranaceus var.mongholicus and identified as rhamnocitrin-3-O-α-L-rhamnopyronosiyl-(1→2)-[6-O-acetyl]-β-D-glucopyranoside(1),kaempferol-3-O-β-D-glucopyranoside(2),quercetin-3-O-β-Dglucopyranoside(3),rhamnetin-3-O-β-D-glucopyranoside(4),rhamnocitrin-3-O-β-D-neohesperidoside(5),methyl-9,12,13-trihydroxyoctadeca-10,15-dienoate(6),methyl-(9S,10R,11E,13R)-9,10,13-trihydroxyoctadec-11-enoate(7),respectively.Different doses of compounds 1 to 5 could reduce the apoptosis rate of HK-2 cells induced by high sugar to varying degrees,compounds 1 to 4 could inhibit the increase of TNF-αand IL-6 levels in HK-2 cells.Conclusion:Among them,compound 1 is a new flavonoid,compounds 6,7 are isolated from Leguminosae for the first time.Compound 1 to 4 can inhibit HK-2 inflammation induced by high sugar.
