摘 要:目的:对山茱萸Cornus officinalis Sieb.et Zucc.中的化学成分及其抗阿尔茨海默病作用进行研究。方法:利用大孔吸附树脂、硅胶、MCI、Toyopearl HW-40C以及半制备液相等多种色谱方法对山茱萸水提物进行分离纯化,使用核磁共振波谱、质谱等技术对化合物进行结构鉴定。采用Aβ_(25-35)诱导PC-12细胞损伤,评价化合物的体外抗阿尔茨海默病作用。结果:从山茱萸水提物中分离得到12个化合物,分别鉴定为:槲皮素-3-O-β-D-葡萄糖醛酸-6″-乙酯(1)、山柰酚-7-O-β-D-葡萄糖苷(2)、2α,3β-双氢杨梅树皮素没食子酸酯(3)、naringenin-7-O-β-(6″-galloylglucoside)(4)、6′-O-vanilloyltachioside(5)、dunnianoside D(6)、3′,5′-di-C-β-glucopyranosylphloretin(7)、4-甲氧基-苯甲酸(8)、对羟基苯甲酸乙酯(9)、对羟基苯丙醇(10)、原儿茶酸(11)、没食子酸-3-甲基醚(12)。体外活性筛选结果表明,化合物3、4、9和12有效缓解Aβ_(25-35)诱导的PC-12细胞损伤。结论:其中,化合物1~5、7~10为首次从该植物中分离得到。化合物3、4、9和12具有潜在的抗阿尔茨海默病作用。Objective:To study the chemical constituents and anti-Alzheimer′s disease effect of Cornus officinalis.Methods:Macroporous adsorption resin,silica gel,MCI,Toyopearl HW-40C,semi-preparative HPLC and other chromatographic methods were used to isolate and purify the compounds of the aqueous extract of Cornus officinalis.The structures were identified by NMR spectroscopy,mass spectrometry and other spectroscopic techniques.The anti-Alzheimer′s disease effect of the compounds was evaluated in vitro using PC-12 cells injury induced by Aβ_(25-35).Results:Twelve compounds were isolated from the aqueow extract of Cornus officinalis and identified as quercetin-3-O-β-D-glucuronide-6″-ethyl ester(1),kaempferol-7-O-β-D-glucoside(2),2α,3β-dihydromyricetin(3),naringenin-7-O-β-(6″-galloylglucoside)(4),6′-O-vanilloyltachioside(5),dunnianoside D(6),3′,5′-di-C-β-glucopyranosylphloretin(7),4-methoxybenzoic acid(8),p-hydroxybenzoic acid ethyl ester(9),p-hydroxyl-benzyl-propanol(10),protocatechuic acid(11),gallic acid-3-methyl ether(12),respectively.In vitro activity screening results indicated that compounds 3,4,9 and 12 could effectively alleviate the damage of PC-12 cells induced by Aβ_(25-35).Conclusion:Among them,compounds 1 to 5 and 7 to 10 are isolated from Cornus officinalis for the first time.Compounds 3,4,9 and 12 show potential anti-Alzheimer′s disease effect.
