酸枣选育新品种遗传多样性的SCoT标记分析

摘  要:目的:利用SCoT分子标记技术分析酸枣选育新品种及酸枣栽培品种的遗传多样性和亲缘关系,为酸枣种质资源的保护、品种改良和优化夯实研究基础。方法:从100个SCoT引物中筛选出条带重复性好、多态性高的引物对供试材料进行PCR扩增,计算不同酸枣品种间的遗传相似系数,用非加权组平均距离法(UPGMA)进行聚类分析。结果:从100个SCoT引物中筛选出17个引物,共扩增出210条片段,每个引物平均产生12.35条片段。多态性条带187条,多态性比率为89.05%。22个酸枣品种间的遗传相似系数为0.561~0.941。聚类分析结果显示,在遗传相似系数为0.72处22个酸枣品种聚为三组。结论:该研究对酸枣种质资源的鉴定保护,分子辅助育种和开发利用提供了理论依据。Objective:To analyze the genetic diversity and genetic relationship of new varieties and cultivated varieties of Ziziphus jujuba var.spinosa germplasm by SCoT molecular marker technology,so as to lay a solid research foundation for the protection,improvement and optimization of the germdasm resources of Ziziphus jujuba var.spinosa.Methods:The primers with good band repeatability and high polymorphism were screened from 100 SCoT primers for PCR amplification of the test materials,and the genetic similarity coefficient between varieties was calculated.The cluster analysis was performed by unweighted average distance method(UPGMA).Results:17 primers were screened out from 100 SCoT primers,a total of 210 fragments were amplified,and each primer produced an average of 12.35 fragments.There were 187 polymorphic bands,and the polymorphism ratio was 89.05%.The genetic similarity coefficients of 22 Ziziphus jujuba var.spinosa germplasms were 0.561 to 0.941.The cluster analysis results showed that at a genetic similarity coefficient of 0.72,22 germplasms were clustered into three groups.Conclusion:This study provides a theoretical basis for identification and protection of germplasm resources,molecular assisted breeding and exploitation of Ziziphus jujuba var.spinosa germplasm resources.
