摘 要:目的:研究川楝Melia toosendan Sieb.et Zucc.果实中的柠檬苦素类化学成分及其细胞毒活性。方法:采用硅胶、ODS、Sephadex LH-20葡聚糖凝胶等多种柱色谱分离技术对川楝果实90%乙醇提取物进行分离纯化,并根据理化性质和波谱数据鉴定化合物结构,利用MTT法测定10个化合物对不同肿瘤细胞的细胞毒活性。结果:从川楝果实的醇提物中分离得到10个柠檬苦素类化合物,分别鉴定为:sendanin(1)、1,7-diacetylsendanin(2)、1-acetylsendanin(3)、7-acetylsendanin(4)、pentaol(5)、meliarachin C(6)、12-O-acetylazedarachin B(7)、azedarachin B(8)、川楝素(9)、meliatoosenin D(10)。10个化合物对5株肿瘤细胞的体外增殖具有不同程度的抑制作用。结论:其中,化合物2~6为首次从该植物中分离得到,且化合物6、9、10对人胃癌细胞MGC803和人结肠癌细胞COLO205表现出较强的细胞毒活性。Objective:To study the chemical constituents of limonins and their cytotoxic activities from the fruits of Melia toosendan.Methods:The 90%ethanol extract from the fruits of Melia toosendan was isolated and purified by silica gel,ODS,Sephadex LH-20 and other column chromatography separation technologies.The structures of the compounds were identified according to physicochemical properties and spectral data.The cytotoxic activity of 10 compounds against different tumor cells was determined by MTT assay.Results:Ten limonin compounds were isolated from the ethanol extract of Melia toosendan and identified as sendanin(1),1,7-diacetylsendanin(2),1-acetylsendanin(3),7-acetylsendanin(4),pentaol(5),meliarachin C(6),12-O-acetylazedarachin B(7),azedarachin B(8),28-deacetylsendanin(9),meliatoosenin D(10),respectively.All compounds had different inhibitory effects on the proliferation of 5 tumor cells in vitro.Conclusion:Among them,compounds 2 to 6 are isolated from Melia toosendan for the first time.Compounds 6,9,10 show strong cytotoxic activities against human gastric cancer cell line MGC803 and human colon cancer cell line COLO205.
