银黄含片对H1N1流感小鼠炎症因子表达的影响研究

摘  要:目的评价银黄含片对H1N1流感小鼠的药效,观察其对H1N1流感小鼠炎症因子表达水平的影响。方法采用流感病毒H1N1CA07滴鼻建立H1N1小鼠模型,奥司他韦组与银黄含片组分别予奥司他韦及银黄含片溶液灌胃7 d。观察小鼠肺指数及病毒指数。采用高通量液相蛋白芯片检测小鼠血清IFN-γ、IL-12p70、IL-17A、IP-10、MIP-1β、ENA-78。结果银黄含片组肺指数较模型组显著降低,模型组小鼠IL-12p70、ENA-78和IL-17A水明显降低(P<0.01),IP-10明显增高(P<0.01)。银黄含片组IFN-γ水平与MIP-1β水平较奥司他韦组均明显降低P<0.05)。结论银黄含片能够明显减少H1N1小鼠的肺损伤,并明显降低IFN-γ与MIP-1β水平,推测银黄含片很可能是通过减少T细胞的趋化,降低流感小鼠的Th1免疫应答,从而起到达到肺保护作用。Objective:To evaluate the efficacy of Yinhuang Buccal Tablet on mice with H1N1 influenza and to explore its effect on inflammatory factors in H1N1 influenza mice.Methods:H1N1 mouse model was established by nasal drip of influenza virus H1N1CA07.The control group and the observation group were given oseltamivir and Yinhuang Buccal Tablet solution respectively by gavage for 7 days.The changes of inflammatory factors includ⁃ing IFN-γ,IL-12p70,IL-17A,IP-10,MIP-1βand ENA-78 in serum of mice were detected by high throughput liquid phase protein chip.Results:The diseased lung index of Yinhuang Buccal Tablet group was significantly lower than that of model group.The IL-12p70,ENA-78 and IL-17A water in model group were significantly lower(P<0.01),and the IP-10 was significantly higher(P<0.01).IFN-γand MIP-1βlevels in the Yinhuang Buccal Tablet group were significantly lower than those of oseltamivir group(P<0.05).Conclusion:Yinhuang Buccal Tablet could significantly reduce lung injury in H1N1 mice and the levels of IFN-γand MIP-1β.It might reduce the Th1 immune response of influenza mice by reducing the chemotaxis of T cells,thus achieving lung protection.
