山楂提取物对高温环境下跑台大鼠T细胞的影响

摘  要:目的 了解山楂提取物干预下高温运动大鼠T淋巴细胞的变化,探究山楂提取物干预对机体免疫的影响。方法 24只6w龄雄性SD大鼠,适应性饲养1w后,随机分为四组,分别为常温运动对照组、高温运动模型组、高温运动补充低剂量山楂提取物组(高温低剂量组)、高温运动补充高剂量山楂提取物组(高温高剂量组)。其中,高温环境的湿球黑球温度(wetbulbglobetemperature,WBGT)系数为33±2℃,高剂量为156mg/kg,低剂量为78mg/kg。跑台倾角15°,转速为14rpm,上、下午各运动30min,每天共1h,共进行4w。每周称量大鼠体质量。4w后,称量胸腺、肝脏和脾脏的质量,计算脏器系数;采用流式细胞仪检测大鼠外周血T淋巴细胞亚群CD3^(+)、CD4^(+)、CD8^(+)T淋巴细胞的百分含量并计算CD4^(+)/CD8^(+)。结果 四组大鼠跑台实验前体质量比较,差异无统计学意义(P>0.05);跑台实验后四组大鼠体质量差异有统计学意义(P<0.05),其中高温低剂量组大鼠体质量最低,增重最少。四组大鼠胸腺系数、肝脏系数、脾脏系数比较,差异均有统计学意义(P<0.05);两两比较显示,高温低剂量组大鼠胸腺系数、肝脏系数高于高温运动组及高温高剂量组;高温低剂量组大鼠脾脏系数高于高温运动组及高温高剂量组;高温高剂量组脾脏系数高于高温运动组,以上差异均有统计学意义(P<0.05)。四组大鼠血液T细胞亚群比较,差异有统计学意义(P<0.05),其中高温高剂量组CD4^(+)、CD8^(+)T淋巴细胞比例最高,CD3^(+)T淋巴细胞、CD4^(+)/CD8^(+)比例较高,表明高温下摄入高剂量山楂提取物,能够提高T淋巴细胞各亚群百分含量。结论 补充山楂提取物可以调节高温运动条件下大鼠T淋巴细胞各亚群的水平,稳定和改善机体免疫系统。Objective To study the changes of T lymphocytes of rats undergoing high-temperature exercise under the intervention of hawthorn extract,and investigate the impact of hawthorn extract intervention on immune function of the body.Methods Twenty-four 6-week-old male SD rats were randomly divided into 4 groups after adaptive feeding for 1 week,which were divided into normal temperature exercise control group,high temperature exercise model group,hawthorn supplement high temperature low dose exercise group,hawthorn supplement high temperature high dose exercise group.Among them,the Wet Bulb Globe Temperature(WBGT)index in a high-temperature environment is 33±2℃.The high dose is 156 mg/kg and the low dose is 78 mg/kg.The running platform has an inclination of 15°and a rotational speed of 14 rpm.Exercise for 30 minutes in the morning and afternoon,a total of 1 hour per day,for a total of 4 weeks.Weigh the body weight of rats every week.After 4 weeks,the weight of thymus,liver and spleen in rats was weighed and the organ index was calculated;The percentage content of blood T lymphocyte subsets CD3^(+),CD4^(+),CD8^(+)T lymphocytes in rats was measured by flow cytometry and the CD4^(+)/CD8^(+)was calculated.Results The difference between the mass of the four groups(P>0.05)and the four groups(P<0.05),with the high temperature and low dose group had the lowest mass and the least weight gain.The differences between thymus coefficient,liver coeficient and spleen coefficient of the four groups were statistically significant(P<0.05).The ison between the differences showed that high temperature and high temperature dose group exceeded high temperature and high temperature dose group;the differences were statistically significant(P<0.05).The blood T cell subsets of the four groups were statistically significant(P<0.05),in which the high-dose group had the highest proportion of CD3^(+),CD4^(+),CD8^(+)and CD4^(+)/CD8^(+),indicating that the high-dose hawthorn extract at high temperature could increase the percentage content of ea
