摘 要:目的运用两样本孟德尔随机化(TSMR)评估新冠肺炎(COVID-19)与带状疱疹之间的关联,探究新冠肺炎是否是引起带状疱疹的因素。方法新冠肺炎和带状疱疹相关的单核苷酸多态性(SNP)数据均来自全基因组关联研究(GWAS)公共数据库,对所有汇总数据进行二次分析,选择与新冠肺炎密切关联的遗传位点作为工具变量,分别以孟德尔随机化线性回归(MR Egger)、中位数加权法、逆方差加权法(IVW)、最大似然比法、线性中位数加权法、逆方差加权径向法、加权众数法等7种方法做两样本孟德尔随机化,以OR值评价新冠肺炎与带状疱疹之间的因果关系。结果上述7种统计学方法得出的正向和反向孟德尔随机化的OR和95%CI值依次为1.82(0.69~4.82)/0.98(0.95~1.01)、1.23(0.90~1.67)/1.00(0.98~1.02)、1.36(0.90~2.05)/1.00(0.98~1.03)、1.23(0.90~1.67)/1.00(0.98~1.02)、1.23(0.91~1.66)/1.00(0.98~1.02)、1.03(0.61~1.75)/1.01(0.98~1.04)、1.38(0.93~2.07)/1.00(0.96~1.04),所有P值均>0.05。两样本孟德尔随机化的异质性检验P=0.39>0.05,水平多效性检验P=0.43>0.05。结论运用两样本孟德尔随机化发现新冠肺炎不具有与带状疱疹相关的遗传证据。Objective To evaluate the association between novel coronavirus pneumonia(COVID-19)and herpes zoster by using two sample Mendelian randomization,so as to explore whether COVID-19 is a factor causing herpes zoster.Methods All the data were collected from the public database of genome-wide association studies(GWAS).The secondary analysis of all the aggregated data was conducted,the genetic loci closely associated with COVID-19were selected as instrumental variables(IVs).TSMR was conducted by seven methods,including Eggerregression in Mendelian randomization,weighted median,inverse variance weighted(IVW)random effects model,maximum likelihood,linear weighted median,IVW radial method and IVW fixed effects model,odds ratio(OR)value was used to evaluate the causal relationship between COVID-19 and the risk of herpes zoster.Results Not more than 7 kinds of statistical methods of forward and reverse Mendelian randomization OR and 95%CI were 1.82(0.69-4.82)/0.98(0.95-1.01),1.23(0.90-1.67)/1.00(0.98-1.02),1.36(0.90-2.04)/1.00(0.98-1.03),1.23(0.90-1.67)/1.00(0.98-1.02),1.23(0.90-1.67)/1.00(0.98-1.02),1.03(0.61-1.75)/1.00(0.98-1.04),1.38(1.93-2.07)/1.00(0.96-1.04),all P values were greater than 0.05.The heterogeneity test results of TSMR P=0.39>0.05,horizontal pleiotropy test P=0.43>0.05.Conclusion There was no genetic evidence of correlation between COVID-19 and herpes zoster by using two-sam-ple Mendelian randomization.
