山莴苣素和山莴苣苦素改善高脂饮食诱导的肥胖小鼠脂代谢及氧化应激水平

摘  要:随机将HFD诱导的肥胖小鼠分为正常饮食组、高脂饮食组、山莴苣素组、山莴苣苦素组、奥利司他组和二甲双胍组,饲喂10周后,利用试剂盒法、实时荧光定量PCR法等方法检测各组肥胖小鼠的各项相关指标,以探讨山莴苣素和山莴苣苦素对HFD诱导的肥胖小鼠脂代谢及氧化应激的影响。结果显示:山莴苣素和山莴苣苦素均能显著缓解HFD造成的小鼠肥胖、降低肝脏TG水平(P<0.01),减少肝脏脂肪空泡、改善肝索病变;能显著升高肝脏T-AOC、SOD水平,显著降低MDA水平。山莴苣苦素能显著升高CAT水平(P<0.05);山莴苣素和山莴苣苦素能显著升高肾脏SOD水平,显著降低MDA水平,山莴苣素能显著升高T-AOC水平(P<0.05)。山莴苣素、山莴苣苦素均可显著下调脂质合成基因Srebp-1c、Fas的mRNA表达(P<0.05),山莴苣苦素可显著上调脂质分解基因Pparα、Cpt-1α的mRNA表达(P<0.05);山莴苣素可显著提升肝脏Nrf2的mRNA表达(P<0.01),山莴苣素和山莴苣苦素均可显著提升肾脏Nrf2的mRNA表达(P<0.001)。因此,山莴苣素和山莴苣苦素可通过影响脂代谢基因和抗氧化基因的表达,改善高脂饮食诱导的肥胖小鼠脂代谢及氧化应激水平。Randomly divide HFD induced obese mice into normal diet group,high-fat diet group,colchicine group,colchicine group,orlistat group,and metformin group.After 10 weeks of feeding,various relevant indicators of each group of obese mice were detected using serum biochemical indicators detection,HE staining method,real-timefluorescence quantitative PCR method,in order to investigate the effects of Lactucin and Lactopicrin on improving lipid metabolism and oxidative stress in HFD-induced obese mice.The results showed that,Lactucin and Lactucopicrin significantly alleviated obesity caused by HFD in mice,reduced hepatic TG levels(P<0.01)and hepatic fat vacuoles,and improved hepatic cord lesions.Lactucin and Lactucopicrin significantly elevated hepatic T-AOC and SOD levels,and significantly lowered MDA levels,Lactucopicrin significantly increased CAT levels(P<0.05);Lactucin and Lactucopicrin significantly increased renal SOD levels and significantly decreased MDA levels,and Lactucin significantly increased T-AOC levels(P<0.05).Both Lactucin and Lactucopicrin significantly down-regulated mRNA expression of lipid synthesis genes Srebp-1c and Fas(P<0.05),and Lactucopicrin significantly up-regulated mRNA expression of lipid catabolic genes Pparαand Cpt-1α(P<0.05).Lactucin significantly enhanced mRNA expression of hepatic Nrf2(P<0.01),and both Lactucin and Lactucopicrin significantly enhanced mRNA expression of renal Nrf2(P<0.001).The conclusion was Lactucin and Lactucopicrin could affect expression of lipid metabolism genes and antioxidant genes and improve lipid metabolism and oxidative stress levels in high-fat diet-induced obese mice.
