摘 要:WRKY家族是高等植物特有且最大的一类转录因子。本研究利用生物信息学技术,在假俭草(Eremochloa ophiuroides(Munro.))全基因组中鉴定92个WRKY家族基因,命名为EoWRKY1~EoWRKY92,分布在9条染色体上。对其序列特性、基因结构、顺式作用元件等进行分析,并基于假俭草相关转录组数据,分析了EoWRKYs在干旱胁迫下的表达模式,最后基于qRT-PCR解析其在干旱胁迫下的瞬时表达特征。结果表明:根据系统发育特征可将EoWRKYs分为三组,GroupⅠ含有完整的WRKY结构域和锌指基序,Group II~III存在结构域和锌指基序丢失和变异现象。EoWRKYs在干旱胁迫下的转录组分析结果表明:在假俭草叶片和根干旱胁迫不同时期,各基因表达量存在差异。qRT-PCR结果表明:干旱处理不同时间后,EoWRKY77,EoWRKY28,EoWRKY30和EoWRKY2在胁迫下表达量显著升高,具有正向调控作用;EoWRKY33,EoWRKY62,EoWRKY9和EoWRKY60表达量下降,这8个基因在干旱胁迫下表达量显著变化,与转录组数据一致。本研究为假俭草WRKY转录因子的进一步功能研究奠定了基础。The WRKY family is the unique and the largest class of transcription factors in higher plants.In this study,92 WRKY family genes,named EoWRKY1~EoWRKY92,were identified in the whole genome of centipedegrass by bioinformatics method.The sequence characteristics,gene structure and cis-regulatory element of EoWRKYs were analyzed,and the expression patterns of EoWRKYs under drought stress were analyzed based on their relevant transcriptome data.qRT-PCR was used to analyze their transient expression under drought stress.The results show that EoWRKYs can be divided into three groups according to their phylogenetic characteristics.Group I contains a complete WRKY domain and a zinc finger motif,which were deleted or changed in Group II and Group III.The results of transcriptome analysis of EoWRKYs under drought stress showed that there were differences in gene expression in leaves and roots during different drought stress stages.qRT-PCR results showed that the expression level of EoWRKY77,EoWRKY28,EoWRKY30 and EoWRKY2 was significantly increased under drought stress,while the expression level of EoWRKY33,EoWRKY62,EoWRKY9 and EoWRKY60 was decreased under drought stress.The expression levels of these 8 genes changed significantly under drought stress,which was consistent with the transcriptome data.This study provides a basis for further functional studies of the WRKY transcription factor in centipedegrass.
