补肺活血胶囊干预慢性阻塞性肺疾病大鼠模型气道炎症及黏液高分泌的作用及机制研究

摘　　要：目的研究补肺活血胶囊对慢性阻塞性肺疾病(简称“慢阻肺”)大鼠模型气道炎症及黏液高分泌的干预作用和机制。方法采用烟熏并气道内滴注脂多糖的方法制备慢阻肺大鼠模型。40只Wistar大鼠随机分为空白对照组、模型组、补肺活血组和茶碱组,空白对照组及模型组予以生理盐水灌胃,补肺活血组和茶碱组分别予补肺活血胶囊和茶碱缓释片溶液灌胃。使用小动物肺功能仪检测大鼠肺功能,苏木精-伊红染色和阿利新蓝-过碘雪芙染色进行形态学分析,酶联免疫吸附实验检测肺泡灌洗液中IL-1β、IL-6、NE、TGF-β、EGF、IL-13和TNF-α的表达,免疫组化法检测大鼠肺组织中黏蛋白5AC(Muc5ac)的表达,免疫印迹法检测Toll样受体4(TLR4)、髓样分化初级应答基因88(MyD88)、NF-κB、Muc5ac、EGFR/p-EGFR、PI3K/p-PI3K、AKT/p-AKT的蛋白表达。结果补肺活血胶囊能够改善慢阻肺模型大鼠FVC、FEV_(0.3)、FEV_(0.3)/FVC和FEF25-75(P<0.05,P<0.01),显著降低肺泡灌洗液中IL-1β、IL-6、NE、TGF-β、EGF、IL-13和TNF-α的含量(P<0.05,P<0.01),减少TLR4、MyD88、ERK1/2、P38MAPK、NF-κB、Muc5ac、p-EGFR、p-PI3K和p-AKT蛋白的表达(P<0.05,P<0.01)。结论补肺活血胶囊可能通过抑制慢阻肺大鼠模型气道炎症及黏液高分泌改善肺功能。Objective To investigate the intervention effect and mechanism of Bufei Huoxue Capsule on airway inflammation and mucus hypersecretion in rats with chronic obstructive pulmonary disease(COPD).Methods The rat model of COPD was established by smoking and intratracheal instillation of lipopolysaccharide.The lung function was measured by small animal lung function meter,and the morphology of airway epithelial cells was detected by hematoxylin-eosinstaining and alicinblue-periodopho staining,the expressions of IL-1β,IL-6,NE,TGF-β,EGF,IL-13 and TNF-αin BALF were detected by enzyme-linked immunosorbent assay,the protein expressions of TLR4,Myd88,NF-κB,Muc5ac,EGFR/p-EGFR,PI3K/p-PI3K,Akt/p-AKT were detected by Western-blot,and the expression of Muc5ac was detected by immunohistochemistry.Results Bufei Huoxue Capsule could improve FVC,FEV_(0.3),FEV_(0.3)/FVC and FEF25-75 in rats with chronic obstructive pulmonary disease(P<0.05,P<0.01)reduce the contents of IL-1β,IL-6,NE,TGF-β,EGF,IL-13 and TNF-αsignificantly(P<0.05,P<0.01);the protein expressions of TLR4,MyD88,ERK1/2,P38MAPK,NF-κB,Muc5ac,p-EGFR,p-PI3K and p-AKT were decreased.(P<0.05,P<0.01).Conclusion Bufei Huoxue Capsule can improve lung injury by inhibiting airway inflammation and mucus hypersecretion in rats with COPD.
