摘 要:目的基于潘多拉测序探索劳力性热射病(EHS)小鼠脑组织非编码小RNAs(sncRNAs)的变化。方法将12只雄性健康C57BL/6J小鼠随机分为对照组(n=6)和模型组(n=6)。对照组小鼠不作处理,模型组小鼠采用恒温恒湿箱内跑步建立EHS模型。EHS模型建立后基于脑湿干重检测和HE染色明确EHS小鼠脑损伤。采用潘多拉测序进行组间差异sncRNAs筛选。结果EHS小鼠脑组织通过PANDORA-Seq共检测2722个sncRNA;与正常对照组比较,以fold change>2、P<0.05为筛选条件,EHS小鼠脑组织共鉴定到53个显著差异sncRNA,如mmu-miR-1843b-5p、mmu-miR-490-3p、mmu-miR-1843a-5p、piR-mmu-7796454和tsRNA-1020-MetCAT等;基于小RNA注释软件SPORTS1.1发掘和注释sncRNAs,提供了长度分布图和种类分布雷达图。sncRNAs种类的差异分析和序列的差异分析结果采用热图和火山图展示。结论sncRNAs失调可能参与EHS小鼠脑损伤的发病机制。Objective To explore the changes of small non-coding RNAs(sncRNAs)in brain tissue of exertional heat stroke(EHS)mice based on Pandora sequencing.Methods Twelve healthy male C57BL/6J mice were randomly divided into control group(n=6)and model group(n=6).The mice in the control group were not treated,and the mice in the model group ran in a constant temperature and humidity box to establish a EHS model.After the EHS model was established,the brain injury of mice was determined based on wet and dry weight detection and HE staining.Differential sncRNAs were screened by Pandora sequencing.Results A total of 2722 sncRNAs were detected by PANDORA-Seq.Compared with normal control group,fold change>2 and P<0.05 were selected as screening conditions,53 sncRNAs with significant differences were identified in brain tissues of mice with EHS,such as mmu-miR-1843b-5p,mmu-miR-490-3p,mmu-miR-1843a-5p,piR-mmu-7796454 and tsRNA-1020-MetCAT.Based on the small RNA annotation software SPORTS1.1 for the discovery and annotation of sncRNAs,the length distribution map and species distribution radar map were provided.The difference analysis results of sncRNA species and sequence were shown by heat map and volcano map.Conclusions Dysregulation of sncRNAs plays a role in the pathogenesis of brain injury in EHS mice.
