鸽源芽孢杆菌的分离鉴定及益生性能评价

摘　　要：研究旨在对来源于健康白羽王鸽肠道中7株芽孢杆菌进行分离鉴定并对其益生功能和安全性进行评价。通过菌落形态,染色特性、生化试验和16S rRNA进行鉴定,对其生长曲线、耐高温能力、耐酸碱能力、耐胆盐能力、产酶能力以及在人工模拟胃肠液环境下的存活率进行益生性能筛选,对其抗生素耐药性、菌株溶血性和小鼠安全试验进行安全性评价。结果共得到7株芽孢杆菌,筛选出3株耐受性、产酶能力和安全性较好的菌株,它们在pH 5~8液体培养基中生长良好;在65℃10 min存活率均在30%以上;在0.07%的胆盐环境下培养24 h后存活率均在84%以上;在模拟胃肠液中培养3 h后的存活率均在42%以上;均可产蛋白酶、α-淀粉酶和纤维素酶;对林可霉素不敏感;均不会产生溶血素;灌胃后的小鼠精神正常,脏器无病理性变化。由此可见,从健康家鸽肠道中筛选得到的3株芽孢杆菌具有良好的益生特性,可作为鸽用微生态制剂的备选菌株。The aim of the study was to isolate and characterize seven strains of Bacillus from the intestinal tract of healthy Columba and to evaluate their probiotic functions and safety.The probiotic performance was screened by identifying its colony morphology,staining characteristics,biochemical tests,16s rRNA,growth curve,heat tolerance,acid-base tolerance,bile salt tolerance,enzyme production capacity,and survival rate in an artificial simulated gastrointestinal fluid environment,and safety evaluation by antibiotic resistance,strain hemolysis,and mouse safety test.The results showed that seven strains of Bacillus were obtained,and three strains with better tolerance,enzyme-producing capacity,probiotic properties and safety were screened out.They grew well in liquid medium at pH 5 to 8;all had survival rates above 30%at 65℃for 10 min;all had survival rates above 84%after 24 h in 0.07%bile salt;all had survival rates above 42%after 3 h in simulated gastrointestinal fluid;all could produce Protease,α-amylase,and cellulase were produced;they were not sensitive to lincomycin;none of them produced hemolysin;the mice after gavage were mentally normal and there were no pathological changes in the organs.In summary,the three Bacillus strains obtained from the intestine of healthy domestic pigeons have good probiotic properties and can be used as alternative strains for microecological preparations.
