摘  要:目的:研究悬浮和贴壁293T细胞在生物反应器内的高密度培养工艺。方法:分别在2个3.5 L固定床生物反应器中加入150 g片状载体,接种贴壁293T细胞和驯化为悬浮的293T细胞,根据葡萄糖消耗情况调整灌流量进行连续灌流培养6天。结果:贴壁293T细胞和悬浮293T细胞均能在固定床生物反应器片状载体上生长,细胞密度最终分别达到2.5×10^(7)cells/mL和2.3×10^(7)cells/mL,6天增殖约50倍。结论:固定床式生物反应可以用于贴壁和悬浮293T细胞的高密度培养。Objective:To investigate the process for high-density cell culture of 293T cells in packed-bed bioreactor.Methods:The adherent 293T cells and suspending 293T cells are inoculated into two 3.5 L packed-bed bioreactors filled with 150 g Fibra-Cel disks.Adjust the perfusion volume according to glucose consumption and perform continuous perfusion culture for 6 days.Results:Both of adherent 293T cells and suspending 293T cells could grow in packed-bed bioreactors Fibra-Cel disks.In the end,the density of cells reached at 2.5×10^(7)cells/mL and 2.3×10^(7)cells/mL for adherent 293T cells and suspending 293T cells,respectively,with approximately increasing 50 folds in six days.Conclusion:The packed-bed bioreactor can be used for high density culture of adherent and suspending 293T cells.
