摘 要:为描述胃癌中circRNA相关的竞争内源性RNA(ceRNA)调控网络,从GEO和TCGA数据库中检索胃癌circRNA,miRNA,mRNA表达谱,识别差异表达的RNA,利用加权基因共表达网络(WGCNA)鉴定关键模块.对关键模块基因做Lasso-Cox回归分析,同时进行生存分析.差异表达分析鉴定了115个circRNA和628个miRNA,WGCNA构建15个circRNA模块和11个mRNA模块.Lasso-Cox分析筛选出4个mRNA.筛选了一系列失调的circRNA,miRNA和mRNA,构建了胃癌中circRNA相关的ceRNA网络.构建的网络有助于加深对circRNA相关调控机制的理解.To describe the regulatory networks of competing endogenous RNA(ceRNA)related with circRNA in gastric cancer,the expression profiles of gastric cancer circRNA,miRNA and mRNA were retrieved from GEO and TCGA databases,and differentially expressed RNAs were identified,key modules were identified using the weighted gene co expression network(WGCNA).Lasso-Cox regression analysis on key module genes was performed,while survival analysis was conduct.Differential expression analysis identified 115 circRNAs and 628 miRNAs,WGCNA constructed 15 circRNA modules and 11 mRNA modules.Lasso Cox analysis identified 4 mRNAs.A series of dysregulated circRNAs,miRNAs,and mRNAs were screened,and a ceRNA network related to circRNA in gastric cancer was constructed.The constructed network can deepen the understanding of circRNA related regulatory mechanisms.
