摘 要:目的观察橘皮素(tangeretin,TAN)对APP/PS1小鼠认知功能,以及脑内小胶质细胞过度激活和炎症因子分泌的影响。方法2.5月龄雄性APP/PS1小鼠,通过口服给予TAN,连续干预16周,观察TAN对缓解APP/PS1认知功能障碍,以及脑内小胶质细胞过度激活和炎症因子分泌的作用。结果通过行为学检测发现TAN明显改善APP/PS1小鼠认知障碍;通过免疫荧光染色,发现TAN显著缓解APP/PS1小鼠脑内小胶质细胞的过度激活;多因子分析检测发现,TAN显著升高小鼠脑内抑炎因子IL-2和IL-5的水平。结论TAN显著减轻APP/PS1小鼠脑内小胶质细胞过度激活以及调节炎症因子分泌,缓解小鼠的认知障碍。Objective To study the effects of tangeretin(TAN)on cognitive performance,microglia over-activation,and inflammatory factors released in APP/PS1 mice.Methods 2.5-month-old male APP/PS1 mice were treated with TAN orally for 16 weeks.The effects of TAN on alleviating cognitive dysfunction of APP/PS1,microglia overactivation,and inflammatory factors released in the brain were observed.Results It was found that TAN significantly alleviated cognitive impairment in APP/PS1 mice by behavioral testing;the results of immunofluorescence staining demonstrated that TAN significantly alleviated microglia over-activation in the brain of APP/PS1 mice;the results of multifactor analysis showed that TAN significantly increased the levels of anti-inflammatory molecules IL-2 and IL-5 in the mouse brain.Conclusion TAN significantly reduces microglia over-activation,regulates the release of inflammatory factors in the brain of APP/PS1 mice,and alleviates cognitive impairment.
