摘 要:目的:使用电泳仪检测PKC/NF-kB信号通路变化探究仿生人工虎骨对于去卵巢小鼠抗骨质疏松的作用机制。方法:研究对象选取SD雌性小鼠30只,小鼠随机分为对照组、手术组及手术+人工虎骨组,对照组进行假手术,不进行卵巢切除,手术组进行卵巢切除手术,手术+人工虎骨组在手术组基础上加用人工虎骨粉灌胃。观察各组小鼠血清ALP,BGP,β-CTX,TNF-α、碱性磷酸酶、抗酒石酸酸性磷酸酶水平,测量各组小鼠股骨生物力学性能并使用电泳仪检测骨组织ALP、BGP、PKC、NF-kB蛋白表达。结果:各组ALP,BGP,β-CTX,TNF-α水平比较差异有统计学意义(P<0.05),手术组ALP,β-CTX,TNF-α水平高于对照组及手术+人工虎骨组(P<0.05);手术组BGP水平低于对照组及手术+人工虎骨组(P<0.05);手术组碱性磷酸酶、抗酒石酸酸性磷酸酶水平高于对照组及手术+人工虎骨组(P<0.05),手术+人工虎骨组与对照组碱性磷酸酶、抗酒石酸酸性磷酸酶水平差异无统计学意义(P>0.05);各组最大应力、最大载荷、断裂载荷、弹性模量比较差异有统计学意义(P<0.05),手术组最大应力、最大载荷、断裂载荷、弹性模量低于对照组及手术+人工虎骨组(P<0.05),手术组BGP蛋白相对表达低于对照组及手术+人工虎骨组,ALP、PKC、NF-kB蛋白相对表达高于对照组及手术+人工虎骨组(P<0.05)。对照组及手术+人工虎骨组比较差异无统计学意义(P>0.05)。结论:仿生人工虎骨有助于提高去卵巢小鼠骨代谢指标,提高小鼠股骨生物力学性能从而实现抗骨质疏松作用,其作用机制可能与PKC/NF-kB信号通路有关。Objective:To explore the anti-osteoporosis mechanism of bionic artificial tiger bone in ovariectomized mice by detecting PKC/NF-kB signaling pathway changes using electrophoresis.Methods:30 SD female mice were randomly divided into control group,operation group and operation+artificial tiger bone group.The control group received sham operation without ovariectomy,and the operation group received ovariectomy.The operation+artificial tiger bone group was given artificial tiger bone meal on the basis of operation group.The serum levels of ALP,BGP,β-CTX,TNF-α,alkaline phosphatase and tartrate-resistant acid phosphatase in each group were observed,the biomechanical properties of femur were measured,and the protein expressions of ALP,BGP,PKC and NF-kB in bone tissue were detected by electrophoresis.Results:The levels of ALP,BGP,β-CTX and TNF-αwere significantly different among all groups(P<0.05).Surgical group ALP,β-CTX,TNF-αthe level was higher than that of the control group and the surgery+artificial tiger bone group(P<0.05);The BGP level in the surgical group was lower than that in the control group and the surgical+artificial tiger bone group(P<0.05);Alkaline phosphatase and tartrate-resistant acid phosphatase in operation group were higher than those in control group and operation+artificial tiger bone group(P<0.05).There was no significant difference in alkaline phosphatase and tartrate-resistant acid phosphatase between operation+artificial tiger bone group and control group(P>0.05).There were significant differences in the maximum stress,maximum load,fracture load and elastic modulus among all groups(P<0.05).The maximum stress,maximum load,fracture load and elastic modulus of operation group were lower than those of control group and operation+artificial tiger bone group(P<0.05).The relative expressions of BGP in the operation group were lower than those in the control group and the operation+artificial tiger bone group,while the relative expressions of ALP,PKC and NF-kB were higher than those in the cont
