摘　　要：IL-31是一种神经免疫因子,可由Th2型淋巴细胞、单核/巨噬细胞、树突状细胞、肥大细胞、嗜酸性粒细胞、嗜碱性粒细胞等细胞分泌;主要通过与IL-31受体A(IL-31RA)和瘤抑素M受体β(OSMRβ)偶联形成的异二聚体受体复合体结合,发挥相应的生物学作用。近年来,对于IL-31及其受体引发相关疾病的机制研究取得了一些进展。研究表明,一方面,IL-31通过激活瞬时受体电位香草酸受体1(TRPV1)、瞬时受体电位受体A1(TRPA1)上调神经元兴奋性等方式参与瘙痒形成;另一方面,通过激活JAK/STAT、PI3K/AKT等信号通路,招募多种细胞因子促进炎症形成。IL-31参与特应性皮炎、结节性痒疹、大疱性类天疱疮等多种皮肤疾病的发生发展。同时针对IL-31受体的生物制剂奈莫利珠单抗,已经批准上市,用于辅助治疗当前治疗无效的特应性皮炎的瘙痒。本文主要围绕IL-31及其受体在皮肤科疾病发生及治疗中取得的相关进展进行综述。IL-31 is a neuroimmune factor secreted by Th2 lymphocytes,monocytes/macrophages,dendritic cells,mast cells,eosinophils,basophils,and other cells.IL-31 makes effects mainly via the heterodimeric receptor composed of IL-31RA and OSMRβ.Recently,there are advances in the research on the mechanism of IL-31 and its receptors causing related diseases.On the one hand,IL-31 up-regulates neuronal excitability by activating TRPV1 and TRPA1 leading to pruritus.On the other hand,IL-31 could recruit a variety of cytokines by activating JAK/STAT,PI3K/AKT,and other signal pathways leading to inflammation.IL-31 is involved in the occurrence and development of various skin diseases such as atopic dermatitis,prurigo nodularis,bullous pemphigoid,and so on.At the same time,a biological agent targeting IL-31 receptors nemolizumab,has been approved for treating the refractory pruritus of atopic dermatitis.This article reviews IL-31 and its receptors in inducing and treating dermatoses.
