摘 要:氟硼二吡咯(BODIPY)类荧光染料是常用的光动力治疗(PDT)/光热治疗(PTT)光敏剂,具备良好的光物理性质。将硝基苯并噁二唑(NBD)与聚乙二醇(PEG)化的BODIPY母核偶联,改造分子的共轭结构得到BDP-1,并自组装成纳米光敏剂BDP-1 NPs,利用粒度电位分析仪分析了BDP-1 NPs的粒径分布与Zeta电位,系统评价了BDP-1 NPs的光学性质、光热转换效率及对HeLa和MCF-7细胞的光暗毒性。结果表明,BDP-1 NPs的粒径约为90.0 nm,具有良好的分散性;680 nm激光(功率密度1.00 W·cm^(-2))照射下,BDP-1、BDP-1 NPs的单线态氧量子产率分别为0.51、0.28;BDP-1 NPs的光热转换效率为62.4%,具有较好的光热治疗效果,光稳定性较好;BDP-1 NPs对HeLa和MCF-7细胞的暗毒性很低,而在光照条件下显示出较好的光毒性。光敏剂BDP-1 NPs有望协同PDT/PTT用于癌症光疗,为光敏剂的开发提供了一定参考。Fluoroboron dipyrrole(BODIPY)fluorescent dye is a common photosensitizer for photodynamic therapy(PDT)/photothermal therapy(PTT),which has good photophysical properties.We coupled nitrobenzoxadiazole(NBD)with PEG-functionalized BODIPY parent nucleus,and modified the conjugate structure of the molecule to obtain BDP-1,which self-assembled into nano-photosensitizer BDP-1 NPs.Moreover,we analyzed the particle size distribution and Zeta potential of BDP-1 NPs by particle size/potential analyzer.Furthermore,we systematically evaluated the optical properties,photothermal conversion efficiency,and photo-dark toxicity of BDP-1 NPs to HeLa and MCF-7 cells.The results show that the particle size of BDP-1 NPs is about 90.0 nm,and it has good dispersibility.Under 680 nm laser irradiation(power density of 1.00 W·cm^(-2)),the singlet oxygen quantum yields of BDP-1 and BDP-1 NPs are 0.51 and 0.28,respectively.The photothermal conversion efficiency of BDP-1 NPs is 62.4%,indicating that BDP-1 NPs has good photothermal therapeutic efficacy and good photostability.BDP-1 NPs shows low dark toxicity to HeLa and MCF-7 cells,while shows good photo toxicity under light conditions.The photosensitizer BDP-1 NPs is expected to be used for cancer phototherapy in coordination with PDT/PTT.This study provides a certain reference for the development of photosensitizers.
