摘  要:本研究成功开发了小麦中铅的免疫层析荧光定量检测卡。该检测卡基于竞争抑制免疫层析原理,使用铕纳米微球作为荧光探针标记抗Pb-EDTA单克隆抗体,构建了一套免疫层析定量分析体系。研究结果显示:铅的灵敏度为0.48 ng/mL,检出限和定量限分别为10.18μg/kg和25.48μg/kg;对认证标准物质的测定回收率为86.6%~110%;精密度结果显示变异系数(CV)均在15%以下。此外,通过层析卡得到的检测结果与ICP-MS方法所得结果间无显著性差异,验证了该方法的准确性与可靠性。This study successfully developed an immunochromatographic fluorescence quantitative detection card for lead in wheat.This detection card is based on the principle of competitive inhibition immunochromatography,using europium nanospheres as fluorescent probes to label anti Pb EDTA monoclonal antibodies,and constructing a quantitative immunochromatographic analysis system.The research results show that the sensitivity of lead is 0.48 ng/mL,and the detection limit and quantification limit are 10.18 and 25.48μg/kg,respectively;the determination recovery rate of certified reference materials is 86.6%-110%;the precision results show that the coefficient of variation(CV)is all below 15%.In addition,there was no significant difference between the detection results obtained by chromatography card and ICP-MS method,which verified the accuracy and reliability of the method.
