摘 要:为建立一种分离人参叶多糖的绿色高效工艺,本试验利用超声辅助水提取人参叶多糖,采用响应面的星点设计(CCD)优化多糖提取产率,利用乙醇沉淀分离出多糖提取物,并检测分析产物中的多糖与半乳糖醛酸含量。结果表明:CCD优化的工艺条件为液料比21.28 mL/g,超声时间58.33 min,超声功率456.67 W,此条件下人参叶多糖提取产率的预测值为2.07%。;在液料比21 mL/g,超声时间60 min,超声功率450 W即修订后的优化参数下,人参叶多糖提取产率为1.95%;提取产物中多糖含量为20.03%,半乳糖醛酸含量为13.79%。该工艺高效节能,可作为人参叶多糖高效分离的方法。In order to establish a green and efficient process for isolating ginseng leaf polysaccharide,in this paper,water based-ultrasonic assisted extraction was used to isolate ginseng leaf polysaccharide.Central complex design(CCD)in response surface methodology was used to optimize the polysaccharide extraction yield.The polysaccharide was isolated by ethanol precipitation,and the content of polysaccharide and galacturonic acid was quantified in the extract.The results showed that the optimized process of CCD was liquid to material ratio of 21.28 mL/g,ultrasound time of 58.33 min and ultrasonic power of 456.67 W,with the predicted yield of ginseng leaf polysaccharide being 2.07%.Under the modified process,i.e.,the liquid to material ratio 21 mL/g,ultrasonic time 60 min and ultrasonic power 450 W,the extraction yield of ginseng leave polysaccharide was 1.95%;the polysaccharide content was 20.03%and galacturonic acid content was 13.79%.This process was efficient and energy saving,and could be used as a method for efficient isolation of ginseng leaf polysaccharide.
