摘 要:文章基于网络药理学探讨功苋止痢散治疗仔猪腹泻的主要化学成分及其作用机理。利用数据库(TCMSP、Herb、OMIM、BIOGPS等)分析功苋止痢散的有效成分及通路。结果显示:共收集到功苋止痢散活性成分49个,其中十大功劳3个、救必应4个、苦参24个、老鹳草6个、铁苋菜12个。根据Degree值,排名前五的成分包括槲皮素、山柰酚、木樨草素、β-谷甾醇和巴马汀;对应靶点262个,关键靶点有转录因子p65(RELA)、丝裂原活化蛋白激酶3(MAPK3)、丝裂原活化蛋白激酶1(MAPK1)、热休克蛋白90(HSP90AA1)、肿瘤坏死因子(TNF)等。通过GO富集分析得到4012个GO条目,细胞成分条目(CC)206个、生物学过程条目(BP)3459个、分子功能条目(MF)347个。KEGG通路富集到230条信号途径,靶点主要作用于高级糖基化终末产物-受体(AGE-RAGE)信号通路、白细胞介素-17(IL-17)信号通路和肿瘤坏死因子(TNF)信号通路。研究表明,功苋止痢散通过槲皮素、山柰酚、木樨草素、β-谷甾醇和巴马汀作用于RELA、MAPK3、MAPK1、HSP90AA1、TNF等靶点,并调控AGE-RAGE、IL-17和TNF信号通路来改善仔猪腹泻。The article aimed to investigate the main chemical components and their mechanism of action of Gong Xian Zhi Li San for the treatment of piglet diarrhea based on network pharmacology.The effective components and pathways of Gong Xian Zhi Li San were determined by databases(TCMSP,Herb,OMIM,BIOGPS,etc.).The results showed that a total of 49 active components were collected,including three Mahonia fortunei(Lindl.)Fedde,four Ilex rotunda Thunb,24 Sophora flavescens Aiton,six Geranium wilfordii Maxim,and 12 Acalypha australis L..According to Degree value,the top five components were quercetin,kaempferol,luteolin,β-sitosterol,and palmatine.There were 262 corresponding targets,and the key targets were RELA,MAPK3,MAPK1,HSP90AA1,TNF,and so on.There were 4012 GO entries,206 CC entries,3459 BP entries,and 347 MF entries by GO enrichment analysis.The KEGG pathway was enriched with 230 signal pathways,and the targets were mainly acting on the AGE-RAGE,IL-17,and TNF signaling pathways.The study indicates that Gong Xian Zhi Li San improve piglet diarrhea by the regulating AGE-RAGE signal pathway,IL-17 signal pathway,and TNF signal pathway through the action of quercetin,kaempferol,luteolin,β-sitosterol,and palmatine on RELA,MAPK3,MAPK1,HSP90AA1,and TNF.
