摘 要:为探明兰花稀缺品种独占春(Cymbidium eburneum Lindl.)玻璃化超低温保存技术中成活率较低的问题,以独占春原球茎为研究对象,研究了玻璃化超低温保存技术对其生理生化特性的影响,并分析了各生理指标之间的相互关系。结果表明:逆境胁迫中,含水量随玻璃化超低温保存进程缓慢降低,卸载处理迅速降低,其含水量是对照处理的79.06%;相对电导率逐渐升高,卸载处理达到最高(86.36%);氧化损伤指标中的丙二醛(MDA)质量摩尔浓度显著升高,玻璃化处理达到最大;蛋白质羰基(PCO)质量摩尔浓度则是先升高后降低再升高,玻璃化处理质量摩尔浓度最低;氧化物质指标中的过氧化氢酶(CAT)活性和抗坏血酸(ASA)质量摩尔浓度变化趋势一致,均先升高后降低,且预培养处理达最大值;超氧化物歧化酶(SOD)活性先降低后升高,呈现双峰变化趋势;活性氧成分中H_(2)O_(2)和·OH质量摩尔浓度在玻璃化处理均达到最大值,且H_(2)O_(2)质量摩尔浓度变化趋势和CAT活性变化趋势在玻璃化超低温保存进程中表现为相互抑制。通过相关性分析,CAT活性和超氧自由基(O·-2)质量摩尔浓度呈极显著正相关,和H_(2)O_(2)质量摩尔浓度呈显著负相关,表明氧化酶物质对活性氧成分具有清除作用。In order to find out the problem of low survival rate in the vitrification-cryopreservation technology of orchid scarce variety Cymbidium eburneum,the effects of vitrification-cryopreservation technology on its physiological and biochemical characteristics were studied with C.eburneum protocorm as the research object,and the relationship between various physiological indexes was analyzed.The results showed that under stress conditions,the water content gradually decreased during the vitrification-cryopreservation process,with a rapid decrease in the unloading treatment,reaching 79.06%of the control treatment.The relative conductivity increased gradually,reaching its highest level in the unloading treatment(86.36%).The malondialdehyde(MDA)mass molar concentration,an indicator of oxidative damage,significantly increased,with the vitrification treatment reaching the maximum level.The protein carbonyl(PCO)mass molar concentration initially increased,then decreased,and then increased again,with the vitrification treatment having the lowest mass molar concentration.The trends in catalase(CAT)activity and ascorbic acid(ASA)mass molar concentration,indicators of antioxidant substances,were consistent,showing an initial increase followed by a decrease,with the pre-culture treatment reaching the maximum value.Superoxide dismutase(SOD)activity initially decreased and then increased,displaying a bimodal trend.The mass molar concentrations of hydrogen peroxide(H_(2)O_(2))and hydroxyl radical(·OH),components of reactive oxygen species,reached their maximum values in the vitrification treatment,and the change trend of H_(2)O_(2)mass molar concentration and the change trend of CAT activity showed mutual inhibition in the process of vitrification-cryopreservation.Correlation analysis revealed a highly significant positive correlation between CAT activity and superoxide free radical mass molar concentration,and a significant negative correlation between CAT activity and H_(2)O_(2)mass molar concentration,indicating the scavenging
