健脾消脂方“异病同治”2型糖尿病与脂代谢紊乱型疾病的作用机制探讨

摘　　要：目的探讨健脾消脂方异病同治2型糖尿病(T2DM)与脂代谢紊乱型疾病的作用机制。方法通过中药系统药理学分析平台(TCMSP)获得健脾消脂方中主要的活性成分及其潜在靶蛋白。利用OMIM、GeneCards和DisGeNET数据库筛选出与T2DM、肥胖(Obesity)和非酒精性脂肪肝(NAFLD)相关的疾病靶点,并通过Cytoscape 3.7.0软件构建“关键靶点蛋白互作”网络图;在DAVID数据库中进行GO富集分析及KEGG通路富集分析。结果检索到健脾消脂方71种活性成分,263个有效靶点,T2DM有效靶点2078个,NAFLD有效靶点2091个,Obesity有效靶点2533个,药物与疾病交集靶点110个。GO与KEGG分析表明健脾消脂方治疗T2DM、NAFLD和Obesity主要涉及的通路有AGE-RAGE糖尿病并发症信号通路、TNF信号通路、IL-17信号通路等;主要涉及的靶点有AKT1、CASP3、TP53、TNF、IL-6、IL-1β。结论健脾消脂方通过健运中州、祛瘀泄浊法异病同治T2DM、NAFLD与Obesity,它的机制可能与改善胰岛素抵抗、炎症反应、脂代谢有关,可广泛应用于T2DM合并脂代谢紊乱型疾病。Objective To investigate the mechanism of“Same Treatment for Different Diseases”of Jianpi Xiaozhi Prescription in treating Type 2 Diabetes(T2DM)and disorders of lipid metabolism.Methods The main active components and potential target proteins of Jianpi Xiaozhi Prescription were obtained from the TCMSP.The related targets of T2DM,Nonalcoholic fatty liver disease(NAFLD),Obesity collected in GeneCards,OMIM and DisGeNET database were be imported into Cytoscape 3.7.0 software to contrast“Protein Interaction of key targets”network diagram.Gene ontology and KEGG pathway enrichment analysis were completed in DAVID Bioinformatics Resources.Result 71 kinds of effective components,263 effective targets of Jianpi Xiaozhi Prescription,2078 effective targets of T2DM,2091 effective targets of NAFLD,2533 effective targets of Obesity and 110 common targets of drugs and diseases were selected.Gene ontology analysis and KEGG pathway enrichment analysis demonstrated that the signal pathways of Jianpi Xiaozhi Prescription in treating T2DM,NAFLD and Obesity mainly included AGE-RAGE signaling pathway in diabetic complications,TNF signaling pathway,IL-17 signaling pathway and targets of AKT1,CASP3,TP53,TNF,IL-6,IL-1β.Conclusions Jianpi Xiaozhi Prescription is used to treat T2DM,NAFLD and Obesity through Jianyun Zhongzhou and Quyu Xiezhuo methods.The mechanism of“Same Treatment for Different Diseases”may be related to the improvement of insulin resistance,inflammatory reaction and lipid metabolism,and it can be widely used in T2DM with disorders of lipid metabolism.
