摘  要:为了研究加味理中汤方(干姜、党参、白术、甘草加茯苓、山药、木香、桂枝、砂仁)对脾虚泄泻小鼠疗效、免疫及肠道菌群的影响,试验将72只小鼠随机分为6组,分别为空白对照组、泄泻模型组、恩诺沙星组及中药低、中、高剂量组,空白对照组小鼠灌胃生理盐水,其余各组小鼠按体重灌胃30 g/kg番泻叶水提物(每天1次,连续9 d)建立脾虚泄泻模型,试验第10天,空白对照组和泄泻模型组小鼠灌胃生理盐水,中药低、中、高剂量组及恩诺沙星组分别按体重灌胃6.5,13,26 g/kg中药提取物及24.15 mg/kg恩诺沙星水溶液,连用3 d。试验期间每日测定小鼠体重、采食量及饮水量,计算日均采食量和日均饮水量并观察腹泻情况和一般状态,统计稀便率、稀便级及腹泻指数;试验结束后检测血清免疫球蛋白(Ig)A、IgG、IgM质量浓度,统计胸腺指数、脾脏指数及结肠粪便中大肠杆菌、肠球菌、乳酸杆菌、双歧杆菌的菌落数。结果表明:治疗前,与空白对照组比较,其余各组小鼠出现脾虚症状,且稀便率、稀便级和腹泻指数均极显著升高(P<0.01),日均采食量及体重均极显著下降(P<0.01),表明脾虚泄泻模型建立成功。治疗结束后,与空白对照组比较,泄泻模型组小鼠脾虚症状持续,胸腺指数及血清IgA、IgG质量浓度均极显著下降(P<0.01),血清IgM质量浓度显著下降(P<0.05),粪便中大肠杆菌、肠球菌菌落数极显著升高(P<0.01)。与泄泻模型组比较,中药各剂量组稀便率、腹泻指数、粪便中大肠杆菌及肠球菌菌落数均极显著降低(P<0.01),且血清IgA、IgG质量浓度,乳酸杆菌菌落数均极显著升高(P<0.01),恩诺沙星组粪便中双歧杆菌菌落数显著升高(P<0.05);中药中、高剂量组体重、脾脏指数、双歧杆菌菌落数、血清IgM质量浓度均极显著升高(P<0.01),中药低剂量组血清IgM质量浓度显著升高(P<0.05)。与恩诺沙星组比较,中药各剂量组In order to study the effects of Jiawei Lizhong Decoction(Rhizoma zingiberis,Codonopsis pilosula,Atractylodes macrocephala,Glycyrrhiza uralensis Fisch,Poria cocos,Rhizoma dioscoreae,Radix aucklandiae,Cinnamomum cassia Pres and Amomum villosum)on the efficacy,immunity,and intestinal flora in mice with spleen deficiency diarrhea,in the experiment,seventy-two mice were randomly divided into six groups,which were blank control group,diarrhea model group,enrofloxacin group and low,medium and high dose groups of traditional Chinese medicine.The mice of the blank control group were gavaged with normal saline;the rest of the mice were gavaged with 30 g/kg of Senna alexandrina leaf extract according to body weight,once a day;and the model of spleen deficiency and diarrhea was established for 9 consecutive days.On the 10th day of the experiment,mice in the blank control group and the diarrhea model group were gavaged with normal saline;mice in the low,medium and high dose groups of traditional Chinese medicine and the enrofloxacin group were gavaged respectively with 6.5,13,and 26 g/kg of traditional Chinese medicine extract and 24.15 mg/kg of enrofloxacin aqueous solution according to body weight;the mice were used for 3 d.During the test period,the body weight,feed intake and water intake of the mice were measured daily,the average daily feed intake and daily water intake were calculated;the diarrhea and general condition were observed;the dilution stool rate,dilution stool grade and diarrhea index were counted;the mass concentration of serum immunoglobulin(Ig)A,IgG and IgM,the thymus index,spleen index,and the colony counts of Escherichia coli,Enterococci,Lactobacillus and Bifidobacterium in the colonic feces were detected.The results showed that before treatment,compared with the blank control group,the remaining groups of mice showed symptoms of spleen deficiency;and the dilute stool rate,dilute stool grade and diarrhea index were all extremely significantly higher(P<0.01);the average daily feed intake and body weight
