摘 要:呼吸道合胞病毒(respiratory syncytial virus, RSV)是全球≤5岁儿童严重下呼吸道感染的主要病原之一,在免疫功能低下人群(尤其是≥65岁老年人)中反复感染往往会导致严重疾病甚至死亡。因此,研发安全、高效的预防性药物和疫苗是预防RSV感染的重要手段。目前,大多数RSV候选疫苗和抗体的主要靶标是RSV表面的保护性抗原F蛋白的融合前构象F蛋白,而RSV G蛋白也是RSV表面的主要保护性抗原之一。G蛋白与细胞表面受体结合后促进RSV感染。在RSV感染过程中,G蛋白通过影响宿主的免疫反应引起RSV相关疾病的发生。其中,在RSV的免疫反应和致病机制中,G蛋白的CX3C趋化因子基序起重要作用。G蛋白在病毒感染过程中的机制及其特征表明,G蛋白可能是疫苗和抗病毒药物研发的重要靶点并具有巨大的研究价值。现就RSV G蛋白相关的细胞表面受体、G蛋白对RSV感染宿主的免疫反应的影响及动物研究中G蛋白致病机制的特征作一概述。Respiratory syncytial virus(RSV) is one of the major causes of severe lower respiratory tract infections in children under 5 years worldwide, and recurrent infections in the elderly and people with compromised immune systems, cardiac and pulmonary systems can lead to severe illness and even death. The development of safe and effective treatments and preventive vaccines is one of the most scientific and economical means to prevent RSV infection. Although the main target of most current RSV vaccine candidates and antibodies is the pre-fusion F protein, the respiratory syncytial virus G protein is also one of the main protective antigens on the surface of the RSV virus. During RSV infection, G protein affects the host's immune response or causes the development of RSV-related diseases through binding to cell surface receptors. Researches prove that the cysteine-X-X-X-cysteine(CX3C) chemokine motif of G protein plays an important role in immune response and pathogenesis, and characterization analysis shows the G protein may be an important target with great potential for the development of vaccines and antiviral drugs. This article reviews the cell surface receptors associated with RSV G protein, the effect of G protein on the immune response of RSV-infected hosts, and the characteristics of the pathogenic mechanism of G protein in animal studies.
