摘 要:目的探讨葛根素改善急性脊髓损伤大鼠神经元功能的作用机制。方法将50只大鼠随机分为假手术组、模型组、葛根素组、PRDM5过表达及葛根素+PRDM5过表达组,每组10只,采用Allen法建立急性脊髓损伤大鼠模型。采用运动功能评分对大鼠后肢运动功能进行评分;肉眼及HE染色观察脊髓组织形态和病理变化;TUNEL染色检测神经元凋亡;免疫荧光染色检测大鼠脊髓组织神经元核抗原(NeuN)、脑源神经营养因子(BDNF)及突触素1(synapsin I)表达;Western blot检测大鼠脊髓组织PRDM5、Wnt1、β-catenin、GSK-3β、p-GSK-3β蛋白表达。结果与模型组比较,葛根素组运动功能评分显著升高,脊髓损伤段淤血面积及结构破坏明显减轻,炎症浸润现象减轻,出现一定量髓鞘再生。大鼠脊髓组织神经元凋亡明显减少,NeuN、BDNF、synapsin I表达明显增加,PRDM5、GSK-3β蛋白表达水平明显降低,Wnt1、p-GSK-3β、β-catenin蛋白表达水平明显升高;且葛根素能够部分逆转PRDM5的作用。结论葛根素可能通过抑制PRDM5表达,进而激活Wnt/β-catenin通路改善急性脊髓损伤大鼠神经元功能。Objective To explore the mechanism of puerarin on neuronal function after acute spinal cord injury in rats.Methods Fifty rats were randomly divided into sham group,model group,puerarin group,PRDM5 group and puerarin+PRDM5 group.The rat model of acute spinal cord injury was established by Allen method.Motor function scores were used to evaluate the motor function of hind limbs of rats.The morphological and pathological changes of the spinal cord were observed by naked eye and HE staining.The neuronal apoptosis was observed by TUNEL staining.The expressions of NeuN,brain-derived neurotrophic factor(BDNF)and synapsin I were detected by immunofluorescence staining.The expressions of PRDM5,Wnt1,β-catenin,GSK-3βand p-GSK-3βwere detected by Western blot.Results Compared with the model group,the motor function score of puerarin group was significantly increased,the congestion area and structural destruction of the injured section of spinal cord were significantly reduced,the inflammatory infiltration phenomenon was alleviated,and a certain amount of myelin regeneration was observed.The neuronal apoptosis in the spinal cord of rats was significantly decreased,the expressions of NeuN,BDNF and synapsin I were significantly increased,the expressions of PRDM5 and GSK-3βwere significantly decreased,and the expressions of Wnt1,p-GSK-3βandβ-catenin were significantly increased.Puerarin can partially reverse the effect of PRDM5.Conclusion Puerarin might improve neuronal function in rats with acute spinal cord injury by inhibiting PRDM5 expression and activating the Wnt/β-catenin pathway.
