摘 要:目的探讨橄榄苦苷对脊髓损伤大鼠神经细胞凋亡及JNK/p38 MAPK信号通路的影响。方法60只8周龄雌性SD大鼠,随机分为假手术组(Sham)、脊髓损伤组(SCI)和橄榄苦苷处理组(Oleuropein),每组20只。脊髓损伤后3 d,进行BBB评分评估运动功能恢复情况;TUNEL实验检测各组大鼠脊髓组织神经细胞凋亡情况;免疫荧光方法与Western blot方法检测各组大鼠脊髓组织磷酸化JNK(p-JNK)、磷酸化p38 MAPK(p-p38 MAPK)蛋白的表达。结果与SCI组相比,橄榄苦苷处理组大鼠BBB评分及斜面试验结果明显升高(P<0.05),脊髓组织神经细胞凋亡率明显降低(P<0.05),脊髓组织p-JNK、p-p38蛋白表达均明显降低(P<0.05)。结论橄榄苦苷能够通过抑制JNK/p38 MAPK信号通路减少神经细胞凋亡继而减轻脊髓损伤。Objective To investigate the effects of oleuropein on apoptosis of nerve cells and JNK/p38 MAPK signaling pathway in rats with spinal cord injury.Methods Sixty 8-week-old female SD rats were randomly divided into sham group(Sham),SCI group(SCI)and oleuropein group(Oleuropein),with 20 rats in each group.On the 3rd day after spinal cord injury,BBB score was performed to evaluate the recovery of motor function.TUNEL test was used to detect apoptosis of nerve cells in rats with spinal cord injury.The expressions of phosphorylated JNK(p-JNK)and phosphorylated p38 MAPK(p-p38)in spinal cord tissues of rats were detected by immunofluorescence and Western blot.Results Compared with SCI group,the results of BBB score of rats in oleuropein group were significantly increased(P<0.05),the apoptosis rate of nerve cells in spinal cord was significantly decreased(P<0.05),and the expressions of p-JNK and p-p38 in spinal cord were significantly decreased(P<0.05).Conclusion Oleuropein can reduce apoptosis of nerve cells by inhibiting JNK/p38 MAPK signaling pathway,thus alleviating spinal cord injury.
