摘 要:目的设计正交试验提取二至丸中的多糖类成分并测定多糖含量,将提取物制成颗粒剂。方法2022年9—12月采用传统的水提醇沉法提取二至丸中的多糖类成分,进行L9(34)正交实验设计,探究料液比、提取温度、提取时间、提取次数对多糖提取率的影响,优选多糖提取物的最佳提取工艺;以葡萄糖为标准,采用苯酚-硫酸法进行样品多糖含量测定;并将提取的多糖提取物进行湿法制粒。结果探究的四个因素对多糖提取率的影响程度为:提取次数>提取时间>提取温度>料液比;最佳提取工艺为:料液比1∶10(g/mL),提取温度95℃,提取时间3h,提取次数3次;所制多糖颗粒剂质量评价结果均符合2020版《中国药典》标准。结论该研究优化了二至丸中多糖类成分的提取工艺,为二至丸的进一步开发利用提供科学依据。Objective To extract the polysaccharides from Erzhi wan by orthogonal experiment,to determine the content of polysaccharides,and to make the polysaccharides into granules.Methods Erzhi wan polysaccharide was extracted by traditional water extraction and alcohol precipitation method from September 2022 to December 2022,and L9(34)orthogonal experimental design was carried out to explore the effects of solid-liquid ratio,extraction temperature,extraction time and extraction times on the extraction rate of Erzhi wan polysaccharide,so as to optimize the extraction process of Erzhi wan polysaccharide.The content of polysaccharide in the sample was determined by phenol sulfuric acid method with glucose as standard.The extracted Erzhi wan polysaccharide extract was granulated by wet method.Results The effects of four factors on the extraction rate of Erzhi wan polysaccharide were as follows:extraction times>extraction time>extraction temperature>solid-liquid ratio.The optimum extraction process was as follows:solid-liquid ratio was 1∶15(g/mL),extraction temperature 95℃,extraction time 3 h,and extraction times 3 times.The quality evaluation results of Erzhi wan polysaccharide granules met the standards of the Chinese Pharmacopoeia 2020.Conclusion This study optimizes the extraction process of Erzhi wan polysaccharide,providing scientific basis for the further development and utilization of Erzhi wan.
