摘 要:目的 应用超高效液相色谱法(UPLC)建立同时测定水红花子药材中花旗松素和槲皮素含量的方法。方法 采用Agilent ZORBAX SB-C18色谱柱,以乙腈-0.1%磷酸水为流动相梯度洗脱,柱温25℃,检测波长290 nm,流速0.2 mL·min^(-1),进样量1μL。结果 建立的分析方法方法学考察良好。花旗松素、槲皮素进样浓度分别在3.258~104.272μg·mL^(-1)、2.983~95.452μg·mL^(-1)内呈良好的线性关系(r~2=0.999 9),平均加样回收率为98.8%、98.8%,RSD值为1.9%、2.2%。结论 本方法简便、快速,可在同一波长下快速测定水红花子中花旗松素及槲皮素的含量,为水红花子药材质量控制提供参考依据。Objective To apply ultra performance liquid chromatography(UPLC) to establish a method for the simultaneous determination of taxifolin and quercetin in Polygoni Orientalis Fructus.Methods The separation was performed on an Agilent ZORBAX SB-C18 column with the gradient elution of acetonitrile-0.1% phosphoric acid in water as the mobile phase,the column temperature was 25 ℃,the detection wavelength was 290 nm,the flow rate was 0.2 mL·min~(-1),and the injection volume was 1 μL.Results The established analytical method has been examined and found to be satisfactory.Taxifolin and quercetin exhibited a good linear relationship within the concentration range of 3.258 to 104.272 μg·mL~(-1) and2.983 to 95.452 μg·mL~(-1),respectively,with a correlation coefficient(r~2= 0.9999).The average recovery rates of the spiked samples were 98.8% and 98.8%,with relative standard deviation(RSD) values of 1.9% and 2.2%,respectively.Conclusion The method is simple,rapid,and can determine the contents of taxifolin and quercetin at the same wavelength,which can provide a reference basis for the quality control of Polygoni Orientalis Fructus.
