运用根形态及黄酮类成分鉴定不同年份黄芪品质

摘  要:为了给黄芪的规范化栽培和合理采收提供依据,对3~8年生6个不同年份黄芪根的形态进行比较,并对根中总黄酮和毛蕊异黄酮葡萄糖苷积累规律进行研究.运用游标卡尺对黄芪根的最粗直径予以测量,通过显微切片染色方法观测其解剖结构并定位黄酮类成分主要位置,利用紫外可见分光光度法和高效液相色谱法对不同年份黄芪总黄酮和毛蕊异黄酮葡萄糖苷含量进行测定.6个不同年份黄芪根直径差异显著,韧皮部是黄芪根黄酮类成分主要储存部位,且5年生黄芪根韧皮部所占比例最高,5年生黄芪根中总黄酮和毛蕊异黄酮葡萄糖苷的含量最高.综合分析认为,黄芪药材的合理采收应以5年生黄芪为宜.Studying on the accumulation of total flavonoids and Calycosin-7-Glucoside in root of A.membranaceus root in 6 different years of 3 to 8,and comparing the morphology of the root to provide a basis for standardized cultivation and rational harvesting of it.The anatomical structure and the main position of flavonoids were observed by microscopic section staining.Measuring the biggest diameter of different samples by vernier calliper,and to measure total flavonoids and Calycosin-7-Glucoside in root of A.membranaceus from 6 different years by utraviolet visible spectrophotometry and high performance liquid chromatography(HPLC).The results showed that the average diameter of the 6 different years was significantly different among one another.The phloem is the main storage site of flavonoids in A.membranaceus roots,and the flavonoids of 5 years root phloem accounts for the highest proportion,and the total flavonoids and Calycosin-7-Glucoside of the 5 years was the highest.According to the above results,it was advisable that the reasonable harvest of A.membranaceus should be focused on the 5 years.
